见过这样的病例。upenn的soft tissue部门是全美兽医院最大的了。他们没见过，估计
也没人见过了。
伤口不是咬伤，和一般的感染也不一样，说the tissue feels like jelly。
怀疑过是chemical burn,但他最开始又只是肿，皮肤好好的。所以应该也不是。
cbc他们已经做了，所有指标都正常，她说这也是很奇怪的现象。
手术时候取了一点肉做biopsy,看是否对确诊有帮助。
现在他们给做了标记，看明天早上会不会持续扩散。然后再讨论怎么办。

Basically,
1. snap-freeze the tissue (presumably unfixed) in Tissue-Tek by liquid
nitrogen.
2. cut sections(I prefer 5-7um). It seems the thinner, the better the
sections stay on the slide. We use plus slide from fisher
3. air-dry at room temperature for 3-5 hours.
4. 2xPBS 3min each to clear tissue-tek
5. Postfix in 2% to 4% PFA in 1xPBS for 5-10 min
6. Wash away PFA by 3xPBS 3 min each
7.PBS+Triton x-100 0.05% 5 min(optional)

you can normalize to the signal to total reads later. I guess it won't make

遵循protocol就行了, 并不需要很牛的人才来管这事
考虑到patient inforamtion and follow up是要非常长的时间,
tissue bank晚建一个月导致的损失都很大, 基本可以说是只争朝夕的事儿
当然有了这么一个库, 能在上面做好translational研究, 那确实是需要
水平很高的biomedical人才. 不过等到找到这样的人才, 才去建tissue bank,
那损失不是一般的大, 实在太可惜了 |

stroma cells， lymphoid cells， endothelial cells.....
即使只看tumor tissue 中的tumor cells， monoclonal 和 heterogeneous 也并不矛盾。
tumor tissue 里的monoclonal 的 tumor cells， 即使有相同的DNA 背景，
但是在transcription 水平，不同的cell populations 的 gene expression patterns
会很不一样。

做short form 的目的是：wild type 和short form是在不同的组织中表达。以前认为
这个gene only expressed in one specific tissue A, but after the lab got the
knock out mice, it has different phenotype. We trying to figure out the
mechanism if these related with the specific tissue A or the gene also
expressed in other tissue but at short form.
我现在是打算把这个short form最长的ORF表达出来看看，但是老板感兴趣的那个组织
我没有检测到，所以还会继续race.

1 现在这个short form的TSS,我测到的是大概90bp的差别，是不是可以说是一个TSS

的，不过我觉得不complete，但是有些phenotype是不需要complete的KO，非要提高KO
效率可以试一下用f/-，我没有试过，或者多打几次TAM，可能更容易变成null。
我对whole body的Cre－er的一个concern是cell autonomous的问题无法address，我认

tissue的细胞分布，看着很简单，想试一下。
我的问题就是：我准备样品的时候，需要control/treated的动物的tissue一样重或者
100%取下来，最后的结果control/treated才可以比较吗？
flow cytometry最后的那个结果,反映的是某种polulation的cell的分布百分比,这个概
念对吗？我们每次杀动物的时候，那个tissue都很100%难取下来，每只动物都只能取下
一部分。
