非常感谢！我也是实在没办法，多加了inhibitor。PMSF也加了。
加了inhibitor的lysis buffer放在冰里遇冷，然后从液氮里拿出tissue，立即
homogenize。。。。
我说的室温过夜是加了6xloading buffer，另外我的buffer里还有2%的SDS，这样也会
降解吗？ |

再做。有什么注意的？我查了一下，有两个方案：
1.放在cryoprotectant里面， 比如DMSO，冻起来。
2. make single cell suspension, 然后保存在ethanol里面。
另外，以前直接冻在-80度的tissue (没有cryoprotectant) 可不可以拿来做flow?
有没有专家给点建议？谢谢。 |

Totally agree. The past year my company interviewed several new PhDs majoring
in tissue engineering and biomaterials - all from very decent groups - but
their Ph. D. works were very poor, no significant contribution to the
fundamental understanding of the field.
Actually, Bob Langer has a more relaistic assessment on its future than most
his students do - see the interview he did with Scientific American for the
Technology Master series. |

was suspended. I don't think there is bone tissue engineering, unless they can
make bone from the real cells.
polyTMC is material the first time I heard, thanks for the information.
Actually we need materials can be used in our printer which require the
material (ink) should maintain their shape after deposition. Can you do it
like this one? If you can make it, you and me can startup a company to compete
with my cur |

我曾经试过多种办法提没有降解（用Bio-Rad or Agilent Bioanalyzer测integrity or
quality (RNA Integrity Number (RIN) by Agilent or RNA Quality Index (RQI)
by Bio-Rad)，不是仅仅用Nanodrop测RNA integrity or quality）的total RNA，然后
做microarray．
1.液氮研磨：RNA quality好，可以做microarray，但是麻烦和太慢（原因你应该知道
）。
２. Glass tissue homogenizer: it didn't work for me to grind tissue in
Trizol in ice bucket with ice, although my tissue was thin and tiny.
Bioanalyzer showed RNA degradation.It's also low through-put.
