第一,food 摄入实验。看看KO的老鼠是不是吃少了。
第二,physical activity test.我想问,除了swim test,还有什么其他的方法能
够测量一个时间段内老鼠的运动量吗
第三, insulin tolerance test (ITT)。我已经和老板说了,我们马上就可以做了。
第四,测量fat mass。
第五,FACS analyze adipocyte progenitor and do some functional assays.

progenitor,至于physical activity, 蛋白质组学,等稍复杂的实验要等到前面的结

FACS analyze adipocyte progenitor and do some functional assays
这种实验没有必要先做的。可能是放到你整个实验的最后那一步。
先是chow diet,检测体重,饮食量,ITT, ogtt or igtt, 血液指标,脂肪含量/体重
,有变化,做个脂肪组织和肝组织的切片看看。有变化再做个能量代谢。
如果chow diet,你的这个ko 小鼠都有变化,那就神了,说明这个基因很强啊。
不管有没有变化,用个High fat diet试试,也许能看出更明显的效果,重复同样的实
验。
如果想做的fancy一点,cross到ob/ob or db/db mice,这个要看你ko mice是什么背景了.
不要用什么STZ之类的。
最后做各种实验,分析机理。
B6j是做metabolism公认的,6n背景,有的reviewer会不认可,当然了B6j是非常容易诱
导成metabolism dysfunction的小鼠了。 |

号在iPS里是根本看不见的。到neuro progenitor stage 有一些,大概10%左右。sort
NCAM以后继续分化,绝大多数才有Tuj1。 |

This question was one of my interested topics. To my understanding,
progenitors cells migrate along radial glia within VZ. Once they decide to
differentiate, they migrate away into destined areas. Migration is just part
of the differentiation program. However there are situations that migration
and maturation can be uncoupled.

erythroid: ter119/cd71, myeloid: Gr1, mac1, B cell lineage: CD19, B220 T
cell lineage: CD4,CD8。这些外周血、骨髓,spleen都做一下.对于block of
differentiation,主要还是FACS和giemsa-wright stain来确定。
还有为了排除microenvironment的影响,还要把KO mice的bm移植给wt mice。至于
osteoclast,最近认为lymphnoid progenitor会呆在osteoblast旁边,并受cxcl12调控
。去年的ASH也有人在osteoblast overexpress beta-catenin可以导致leukemia的

邓用的是MEF,把一个分化的细胞推回原始状态,但是如果我们看cacrinogenesis,很

就是说CD34阳性细胞会迅速转化成阴性细胞,用标准培养体系培养一周后可以从90%降
到10-20%。 Bone marrow 稍微慢一点,买这个细胞一般很贵,自己分离的话其实也不
难,但是没有经验自己摸索可能得费些时间。 |

化过培养体系,可以维持CD34+细胞两周左右的时间,然后纯度就会从刚分离的90%以上

经实现small molecule reprogramming了,human也已早有单一OCT4实现Neural
progenitor cells reprogramming的案例。
如果是应用研究,那么现在主流的reprogramming都采用非整合方法了,质粒和mRNA都
很成熟方便,也已经都商业化了。用Retro和Lenti做出来的绝大多数iPS也被认为是质
量较差的细胞株,实用价值很低。 |

目前尚无定论,现有的in vitro model的数据似乎并不支持但也无法完全排除这种可能
。然而entry inhibitor确实可以有效阻止viral spread in vitro and in vivo,那么
合理的猜测就是即便存在细胞间的传播,也很可能是经由受体介导的。
hepatocyte的再生比较特殊,即便是成熟的肝细胞本身仍然具有很强的增殖潜力,此外
也存在liver progenitor cells。但有证据表明,HBV只感染高度分化的肝细胞,原因
之一就是NTCP只在分化的肝细胞中高表达。理论上讲,如果能激活肝细胞再生,同时抑
制病毒复制尤其是new infection,是有可能通过“稀释效应”最终清除viral
reservoir的,当然这仍然需要相当长的时间彻底清除病毒,很难像HCV DAA治疗那么强

are many types of normal cells that are not differentiated at all, such as
progenitor cells or stem cells, but, they are not cancerous. Also, certain
cancers are sometimes well differentiated. But, cancer cells must have the
ability to proliferate unlimitedly or immortalize. Ki67 or other markers
may not be good methods to assess the proliferation in some cancers.

分到大概2千到5千个细胞(FACS分捡),所以没法做western,最后的细胞蛋白样可能会
几个老鼠分出来细胞POOL在一起,但是还是不够,请问还有什么敏感的蛋白质检测方法
去检查蛋白(用抗体)? 因为样品量实在太少了,多谢各位指教。谢谢。 |
