最近nature有一篇文章用transposon+barcode说即使是transplant后体内绝大多数造血
过程也是由progenitor cell而非stem cell负责的,当然他们主要是用老鼠,我怀疑在
人里做时间长了之后会不会看到stem cell的克隆。这个strategy对其它系统也应该适
用。
PS:我觉得很多组织器官因为turnover慢其实很少涉及到tissue homeostasis的概念,
血液系统,肠道,皮肤这种turnover速度很快的系统才涉及到homeostasis,所以只有
在challenge这些homeostasis比较慢的组织器官时才能找到特定细胞的功能

这方法够牛B, 不过结论其实并不算太出奇啊。 跟我之前探讨的一样,homeostasis or
repair of mild damage 由progenitor负责,repair of catastrophic damage由stem
cell负责。

实际上我觉得Pax7细胞就不应该叫做stem cell,
叫个其他名字,比如progenitor cell。
大家都管自己的细胞叫干细胞,实际上只不过是炒作概念,
做development的都说自己是做的干细胞了。

TA cells是个stem cell 的动力学问题. 对于fast turnover tisue以及slow turnover
tissue,我认为其dynamic应该是不同的。fast turnover tissue 来说,quiescent
stem cell-->proliferating TA cells——》committed progenitors_-->terminal
differentiated cells 这样的过程有利于stem cell快速扩增。但对于很多slow
turnover tissue, 比如,肌肉,骨,这样的TA population在生理状况下是检测不到的
。当然对slow turnover tissue的 stem cell dynamic的研究本来就很少。

总觉得说一个pathway控制organ size 实在太简化了点。
换个角度看,发育的过程就是embryonic progenitor cell逐渐specify, restrict成为
lineage specific stem cell或者消失的过程。看看Hippo pathway是如何调控stem
cell的形成,特化应该会很有意思。

这个问题问得好。专门对干细胞衰老的研究好像没有。因为干细胞的传统定义就是life
long maintenance.理想得被定义为永远具有更新能力。自然不具有衰老问题。
我也在琢磨这个问题,如果在病理状况下,比如我contituional activate某signal
pathway 来不停的激活qienscent stem cell,那他们会不会最终衰竭。再比如糖尿病的
创口不愈合是否就是由于干细胞的提早衰老导致?
不对称分裂的行为在干细胞里研究很多。如楼上所说,在低等生物里常见到
asymmetrical division. 以此来维持stem cell population的总数问题。但在
mammalian里,不对称和对称分裂都有。比如复层化前的上皮干细胞的不对称分裂就类
似与
drosophila的 germ cell。symmetrical也不少 好像satellite cell就是, 对称分裂在
群体水平上,有的give rise to two progenitor, 有的give rise to two stem cell.

zika导致microcephaly, CDC已经conclude了。不过,主要的证据——两个流行病学研
究sample size很小,证据还不够强。

哥伦比亚一个儿科大夫觉得她的病人里多了两个小脑袋(microcephaly), 就问孩子他
妈,有没有被蚊子咬过,觉得不舒服。在那个鬼地方,谁不被蚊子咬。于是她就说这跟
zika有关。于是就上了报。美国人看到了吓坏了。不是害怕发烧呕吐,而是害怕小脑袋
。马上调查zika的分布。发现3/4的美国都有,北到Michigan。媒体用扩散(spread)这
个词,听上去像是蚊子几天就从南美飞到Michigan。
zika不仅美洲有,非洲和亚洲也有。跟小脑袋的关系只在南美局部发现。非洲和亚洲蚊
子一样多,一样厉害。但是,没有发现那个关系。
zika不是突然从天上掉下来的,它和人类的关系历史悠久。小脑袋突然增多,可zika年
年都那样。那个CDC文章引用的主要数据不是研究论文,而是巴西国家统计数字。作者
都没敢说microcephaly的增多跟zika outbreak有关,只是说recognition of Zika
