Communication), 关于in vitro formin如何感应力并调控actin生长速度。不知道你看
到没有。in vivo 也有很大进展。Sheetz组现在能够直接in vivo 测量 focal
adhesion protein的end-to-end distance. 其中talin的工作去年已经发表在PLoS
Biology上。 今年会有一系列别的蛋白的结果出来。会非常有趣。
in vitro adhesion protein如何感受机械力,如何unfold, 如何因此和别的signaling
protein相互作用把机械力信息转化成生化反应信号传递,今年会有一系列工作出来,
包括talin-vinculin, alpha-actenin - vinculin, 等。
上面这些全部都是直接单分子测量,和模拟无关。 我觉得你说得挺对的。这个方向似

少有组织学,生理学的实验。当然最后总会用一个行为学的实验来验证一下genetic
phenotypye,剩下的就是用生化解释pathway。感觉CELL老多都是这样的PAPER。起点都
是一个IN VIVO或者primary cell tissue, 中间做着做着,就看见293T cell, 然后
FLAG-TAG。我现在一看见有293T cell的文章,感觉都头大。那些生化PATHWAY用293T解
出来的,到底有多少是能真实的反应in vivo的生理。 |

有尽量多的结构信息。但结构即不是必要条件也不是充分条件。至于离最后做出新药还
有十万八千里。说什么解出结构就看到新药的曙光,征服什么什么疾病,那是纯粹胡扯。
就结构本身,当然in vivo的结构是最有用,但是目前没有有效的手段得到对做新药有
用的结构。那么解晶体还是目前最有效的也是最成功的手段。所以一说到解结构,那么
大家首先默认的就是X光衍射解晶体。至于晶体状态和in vivo状态下结构的差异,确实
是个conern,但是在很多晶体结构还渴望不可及的情况下,基本上还不需要太concern
这个问题。其他手段也有NMR,包括这个电镜,有不错的例子,也有他们的优势,但总
体上和晶体结构提供的information差远了。至于这个4.5A的电镜结构,对做药基本上

降低,那么这就是一个promising的现象,可以围绕这个来开展实验。
至于体外分化的结果, 我觉得除非敲掉的是t-bet,gata3这种基因,否则很难有很显
著的差异,我认识一个人做Th17,他体外分化后测Il17,有时候好一点大概是30% vs
40%, 有时候差一点就基本没差别了,最后还是照样发NI,关键是in vivo的差异很明
显。
仅供参考。
另外,就是感觉这几年这种题材的文章越来越难发NI这种级别的杂志了,基本没有
novelty了,大家觉得呢? |

vitro筛选的效力惊人,只要有足够的化合物,in vitro活性都可以得到。
真正的瓶颈在in vivo筛选,这里面的最大问题的动物模型,现在最常用的是大鼠和小
鼠。这些动物in vivo和人类的相关性、预测性等完全没有办法,动物模型本身的病理
状态和人疾病的相关性等。如果哪一天猴子模型可以很普遍的应用,可能这些预测性和
相关性会好很多。

小鼠被广泛用于模拟人类代谢、疾病和药物应答,是医学研究中的一个基本工具。然而
斯坦福大学的研究指出,人类和小鼠的基因表达存在着惊人的差异,不论是蛋白编码基
因还是非编码基因。这项研究发表在十一月十七日的美国国家科学院院刊PNAS杂志上。
相信在未来的不久对大鼠基因表达也会有类似的研究,这样应该可以比较好的解释为什
么动物模型的相关性和预测性是如此之差。

异元细胞在子宫里融合, 一个人的不同器官有不同遗传信息,现在不太清楚这个比例
占多少, 如果不是小概率事件, 对实验结果的影响应该会很大,
当然,我之前的研究对象是植物, 不太清楚在植物界中是否有相似的情况
我是转行一族, 之前做生化实验还可以, 但in vivo的结果差异太大, 而且不只是我
一个人, 而是整个群体, 经过2~3年的经历, 使我认识到凭自己的微薄之力,只能苟
延残喘, 不弄虚作假, 很难生存, 高质量的研究没有in vivo怎么能行, 这么多基
因, 找到一个好基因除了碰运气, 比海底捞针还难, 我可以选择继续前行, 但亡羊
补牢,为时未晚, 苦海无边,回头是岸; 与其回国做一个正教授, 整体顶着卖白粉
的压力, 干着卖白菜的买卖, 我感觉凭我的能力,找对方向应该比这个更强。

是没有谁用fluorescent microscopy做了(resolution也打不到啊)。最近的in vivo
super-resolution也只是证明了nucleosomal clutch(发在cell上,大家自己找),没
有证明bead-on-a-string啊?哦,对了,biochemistry method, MNase digestion, 再
提取DNA, gel。OK。但是这个MNase digestion的chromatin不也是提取的吗?
避而不谈这个问题了。我实在想不出有什么其他biochemical method 去印证我观测到
的结构了。

2016年生物领域的 {韩春雨事件} 已经进行了5个多月了。
由于无法被同行重复,韩春雨的NgAgo基因编辑工具,已经被世界各地做基因
编辑的科研人员们放弃了。同行们会接着使用CRISPR这个很可靠的基因编辑
工具。
虽然基因编辑行业现在已经放弃了韩春雨的NgAgo,韩春雨还是对世界科研界
造成了不小的影响。
那么,这个事件的影响有多大呢?
据Addgene 2016年8月份统计,已有近30个国家的大约400个实验室从
Addgene拿到了韩春雨的NgAgo 质粒。而中国的实验室,很多是直接从韩春雨

问题是各种高分子的生物兼容性问题。
或者把生物大分子当作纯粹根据官能团可推测化学性质的高分子。
我对第一种有些了解,由於生物现象的复杂性,简单的分子链官能团变化而导致的不同化
学性质的高分子,在生物体内,即所谓in vivo, 对蛋白质,细胞的反应大同小异。现在
最讽刺的就是in vitro 的结果常常和in vivo相反。所以除非生物上游有突破,否则只是
盲人摸象。
第二种我也有同样的想法。

applications, no matter how well you cap it. So the issue is the material of
the nanoparticle core, not the ligands (arms) aroud it. People have been able
to make sort of biocompatible nanoparticles by incorpting PEG and peptides
onto nanoparticle surface, but still there is no way to make them intact in
physilogical conditions for a long time. Nanoparticles with gold or silica
cores are more likely to be used for real in vivo |
