你说的非常对，PCR来检测DNA repair非常罕见
我要解释一下，
我压根不是做DNA damage/repair的，我有兴趣的一段疾病相关的序列repetitive
simple sequence，根据各种原因我相当怀疑是和DNA damage/repair相关的。我的
hypothesis是：这段疾病相关的repetitive序列或许更加vulnerable to 外界的DNA
damage， 或者DNA repair的修复能力跟其他普通序列不同，从而能为疾病机制提供新
的发现
我看了大部分DNA damage/repair领域的文章，我感觉大部分的工作都是global的比如
测量一下DNA损伤的marker比如H2AZ或者H3k56ac的level。但我的兴趣一定是需要精确
到DNA-specific或者gene-specific的
能精确到sequence-specific的我能想到的也就是PCR了

SV的问题是，有不同种类deletion，insertion，duplication, CNV, translocation,
inversion, segmental duplication...
而且有不同的size，小的1bp，大的好几MB
这两个维度都是SNP研究里不曾遇到的
如果你要问一个human being有多少SV？ 我觉得目前都无法回答。因为目前的研究大部
分还是基于short reads的针对基因组的uniq sequence。真正SV hotspot是repetitive
region，但我们无法研究。当然，对于NA12828，人们做了大量研究，really high-
depth seq, Pacbio long reads，或许对这个样本的总结可以给你答案
所以不管是NGS技术，还是后续的计算，SV都是基因组学最大的难关；而难关中的难关
就是那些repetitive seq。
2. 这些SVs是不是对于以后个性化治疗什么的极为重要

信息量的情况。。。（当然，cantor 也可能早就知道，但是作为高维生物玩弄一下低
维生物，所以 cantor 故意不说）。
这样这个 permutation (with repetition) on infinite length sequence，直接生生
造出了许许多多以前从来没看到过的“不可计算数”，原因是那种数的香浓信息量无穷
大。。。（顺便说一句，sqrt(2) 是可计算数，虽然是无限不循环小数，因为其携带的
信息量不是无穷大，或者说有有限长度的生成规则产生，即使序列长度是无穷长且没有
肉眼直接看得见循环 pattern，这不等于没有理论上的 pattern。。。当然这又要回到
哥德巴赫猜想，是不是所有的 pattern 都基于同余循环的问题）。
这种不可计算数多如牛毛的程度，到不管是 dedekind cut 随便来一刀，还是 cauchy
sequence 随处震三下，其结果是数学上严格精确的 100% 的概率出现的是不可计算数

计算数是玄学，不可计算数是 cantor diagonal argument 的无限序列上的排列树 (
permutation w/ repetition) 搞出来的，在马工实践上球用没有。
而这一切最终归结于测度论的必须的地位。。。而测度论的必须的地位，是因为理论纯
数学家认为绝对无二义性的集合论是数学的基础，最终导致无法避免用无限个没有大小
的点拼出整个时空的测度论攻略大全的问题。
但还是数学家目前还是证不出哥德巴赫猜想。 |
