purify B cells from PBMCs will be better than PBMCs.
As for human B cell line, I think most of them should be from B cell
lymphoma. First, you do not know whether they secrete IgG1, second you do
not know how their DNA changed.
I do not know to what extend the gene of IgG1 heavy chain preserved in
different B cell clone, but definitely this is what you must take into
consideration.

nt mice you use, you need less cells transplanted. I wonder if you just use
HELA cells, do you get the same result? then, there is a small number of CSC
in HELA cell culture too?
And what does this mean regarding to clinical practice?
4. More adencarcinomas are more due to mutations (Kras, EGFR), less due to
promoters. I tried different promoters, all leads to adenocarcinomas. I am
switching
to other mutations.

Sure, some cells in tumor popolation will be resistant to therapy. Are they
, however, CSC? For example, some tumor cells finally will be resistant to
target therapy and survive. These cells usually develop novel mutations in
the genes that target therapy acts on. In this case, these cells are
propably not CSC.

evidence for CSC so far are mostly correlative. For me the key experiment
would be:
If you selectively deplete the so-called "cancer initiating cells" without
affecting the bulk of the tumor, will you see the regression of tumor?

The atmosphere in this group is not healthy. And problem like this generally
seeds from lacking proper managerial skills of the top, meaning: you can do
nothing and may only take heat. As a rotation student you have a long way
to go to get to the PhD level and during this process you need lots of
SINCERE help from your colleagues, which i just don't see it happen in this
group.

发帖数: 287 | 28 我觉得你这个实验比较适合用 cell sorting 来做。取单细胞做 Single cell PCR 的
时候细胞周围溶液容易被周围破裂了的细胞污染，假阳性率很高，而且样品也不容易有
代表性。做 cell sorting 的话不同颜色的细胞收集下来多洗一洗，破损细胞的 RNA
去除得干净一些，还能在一个很大的样本量上比较不同颜色细胞的 RNA 表达，听起来
更靠谱一点。
我在培养细胞里做 Single Cell PCR。把 293 或者 culture neuron 用酶消化，用
PBS 稀释得非常淡，1：1000-3000吧，在显微镜下找到单细胞，用一微升的枪把细胞吸
进枪头，然后就是按照 cDNA kit 的配方直接做 cDNA，也没用特殊的步骤破细胞。我
用的 ABI 的 kit，做出来的 cDNA 有 10 微升，全部用来做 PCR。 |

我做了一个toxin，之前他是没有毒性的，然后我想注射到组织内，想用cancer特有的
蛋白的antibody连上，然后利用在局部区间高溶度，是的toxin进入细胞内，也就是穿
刺，最后将保护toxin的外壳去掉，利用toxin的来杀死cancer cell.所以我的问题是：

with telomerase, it can proliferate indefinitely-immortalized, but still

CSC也是属于Cancer的吧？或者是良性的？一旦生长上面出现无法控制的情况，就变成
恶的了？另外，良性的肿瘤有没有tumor stem cell，原来遇见过子宫肌瘤，西瓜那样
大，里面是否也存在tumor stem cell呢？
CSC现在争论是否已经达成一致了？还是Cancer Initiating Cell?
不过从Leukemia来看，CSC应该是存在的，尽管不如HSC具有那样明确的hierarcy；对于
实体肿瘤，很多认为CSC也是存在的，尤其是今年Cell上发表Barret食管癌相关的那
篇paper也提示存在这么一个状态；而从lymphoma的研究来看，CSC好像还没有明确的找到，一方面可能因为lymphoma本身分类比较复杂，HL里面有人认为其中的小细胞可能是CSC，但也有人认为
患者体内CD20+的细胞为HL的CSC；而对于NHL，可能发展到leukemia的时候，会有CSC，
若只停留在lymphoma的状态，好像也还没有CSC的报道。其他器官如breast、prostate
、liver、brain等似乎都有很多CSC的报道。

文献上说10Gy single dose, about 100% cell death.
同样的cell line，CRO 做了 upto 50Gy/dose, 1 dose/day. After 3 days, the
cells are still fine. 这是不是太违法常理了？ 我强烈怀疑他们机器坏了。请有经
验的同行指点。 |

chimera，
或者是ES cell contribution比较少，需要筛选很多后代才能得到germline
transmission,
公老鼠breeding就比母老鼠要有优势。
用来注射ES cell 的blastocysts 的性别比是 50：50，但是如果ES cell
contribution比较多的话，
就很可能把本来应该发育成母老鼠的“受体” blastocysts “反转” 成公老鼠。所以
得到chimera是公老鼠
的概率总是比母老鼠要大。
选xy ES cell唯一的drawback大概就是如果想target x-chromosome gene,得到正确克
隆的概率要小些。 |

am thinking, the less passaging the better...otherwise one might be running
a risk of picking the cells from a same clone multiple times; as well, the

cytometry没问题但做cofocal isotype control 的背景很强.目前为止blocking 效果
最好的就是老鼠血清(用的抗体是monoclonal mouse antibody directly conjugated
with Alexa Fluor),但问题是背景没了,信号也弱了好多.这个实验组和isocontrol 比
起来亮不了多少.现在打算用1抗加2抗做,一般是用和二抗同种的血清来做blocking,但
听之前染过这个蛋白的人说,二抗血清对B cell Fc没什么作用.我的问题如下:
1. 不知道这种情况下能不能先做Fc blocking再用二抗血清block?
2.Caltac Laboratories (now belong to Invitrogen)原来有个抗CD32,但不连接荧光
素的已经不卖了,而且那个也是从老鼠养出来的.请问有什么非老鼠的抗体或血清适合做
human B cell FcR blocking?
