fuction myfun(dataRange as Range). In this function the selected cell
matrix will be stored in an data array. I used to use Range.CurrentArray or
Range.Value2 to store the data into an assigned array variable successfully.
However, it requires the cells contain no formula. If the data in the cells
are obtained by a formula, everything screws up.

transdifferentiation. A few weeks later, there were 2 science paper did
transdifferentiation. When Cell launched Stem Cell, she was in the editor

population, and cancer can be derived by different cell lineages, at least
in some types of cancer.
Plus I don't think people in cancer stem cell field thought the "cancer stem
cell" have the ability to differentiate into multiple cell types. They are
more like cancer initiating cells per se.

ns )of cells. The bias of sequencing or SNP would lead to identify the most
obvious low hanging fruit, such as KRAS, pten, p53 etc.
Apparently when single-cell level sequencing becomes widely available, iden
tifying subpopulations with genetic or epigenetic characteristics will be i

Lineage reprogramming is definitely very promising. As mentioned in another
post, it has been showed that fibroblast can be transformed to neuron
with only 3 TFs.
At the same time, direct programming from ES/iPS cells to somatic cells are
few. And I was not be able to find any publications. So was wondering if
someone can help me with some papers about direct programming from ES/iPS
cells to somatic cells with TFs, not stepwise extracellular signals.

difficult to freeze down. Most cells were dead after frozen with 10% DMSO
in serum. Other cell lines are pretty normal. So anyone can give
suggestion on this problem?

I guess the simple experiment is to give cultured cell line radiation (mimic
those of cell phone) for months or longer and see any effect. I guess some
people did it but did not find anything interesting.

cell is attached, I will just do PI stain. You can also count the cell
directly with trypan blue etc.
However, I can tell you the reviewer is been difficult. I bet you already
count the cells and your compounds do inhibits cell growth....I haven't seen
a single compound that could lower the mitochondria function so much that
will affect the MTS assay if it is not directly reflective toward cell
numbers. |

但是我强调得是“几乎没人听说过”,我们实验室将近20个人,做得是细胞/遗传/生
化。。。18个人没听说。。。这也算侧面证明cell research影响力不行吧?毕竟领域
接近啊。
而且不能完全用“新杂志”来解释,很多cell和nature子刊得新杂志因子也不是那么高
,但是在领域里影响还不错。
大家就像一个身边得例子吧,你的研究文章的重要文章那篇综述或者research文章是引
用cell research上得?
当然我不是说cell research就不好,我知道很多国内得实验室被cns screw 得文章就
发到cell research上面 |

ES细胞分为naive和prime两个stage
1)naive stage:ES colony在形态上看起来更“致密”,呈三维结构,对胰酶不敏感(
即胰酶消化不会诱导其凋亡)。对小鼠ES而言,naive的ES来源于inner cell mass(ICM
)。此stage的ES self-renewal更迅速,population double大概只需要12-14小时。对
于其self-renewal的维持需要LIF,stat3,BMP4等。雌性小鼠naive ES的另一个显著特
点是X chromosome reactivation,而在其他类型的细胞中有一条X chromosome是失活的

protocol可能需要re-consider了。比如上面那个protocol,neonatal mice limb
muscle.分离出来的很有可能是混杂了satellite cell以及committed myoblast cell的
mixture,而到底是哪种细胞长起来就很难说了。
adult的myoblast cell culture好像没有吧。 我没有仔细查过文献,只是大概的尝试
过几次,觉得从adult muscle fiber分离出来的细胞体外很难长。 |

工作,说到cell,非生物专业的人才很多不熟悉这个杂志,他们充其量认为cell是生物
学领域的顶级期刊,而nature和Sceince是全领域的顶尖期刊,比cell高一档。
后来仔细想想,他们非生物人士说的也对,毕竟nature和Science是多领域杂志,cell
也就和Nature有关生物医学的子刊(20以上的)一个档次吧!
把cell抬到nature和Science等同的地步,有点降低NS的档次! |

把买来的competent cell 摇起来一些,然后做competent cell,但是效率似乎非常的
低,不知道版上有没有人有过类似的经验。我之前一直自己做competent cell (不同
菌株),效率非常非常的高。
是不是要传代几次啊? |

如果multipotent的标准都不要了,hierarchy的标准也不要了,那已经没有探讨stem
cell的意义了。就像在liver和pancreas里边一样,每个cell都可以是stem cell, all
cells are born equal! ———— 这显然歪曲了学科的本质。

repeat)位点
大概三个月前测的是这个位点是homozygous有6个repeat
然后最近重新解冻另外一个stock,start cell culture,测量同样的位点,居然成了
heterozygous,一半仍然6个repeat,另外一半成了10个repeat;反复做了很多遍,换
了所有的reagent生怕是contamination,但结果还是一样。
问了lab technitian,这个neuroblastoma在我们实验室都是一样的来源,他也怀疑这
个repeat是不是dynamics的
三个月前的cell line的passage不记得了
最近这个cell line的passage大概是p10
我想问下,cancer cell line的genome microsatellite难道真的会传代传着传着就发
生了变化?如此不稳定? |

献好像T细胞都是用1640),但lab过去的cancer cell 都是用DMEM (corning 10-013-
CV),如果我现在用1640,就会导致实验condition改变了,跟原来的cancer cell的实
验不好比较。
但我没看到用DMEM来养T细胞的,想问问我能不能用DMEM (加上IL-2之类T cell需要的
因子。。)来做我的co culture?如果不能,为什么?谢谢! |
