因为靠热运动组装第一个蛋白质是困难的，所以有观点认为第一个蛋白质是根据某个自

在俺看来，这是先有蛋还是先有鸡的问题。蛋白质好比蛋，基因好比下蛋的鸡。如果自
然界糊出一个蛋很困难，那糊出一个下那个蛋的鸡就更困难。
事实上，每三个DNA碱基对决定一个氨基酸，加上基因的头码三个，尾码三个，对应一

很大程度上这一步的效率取决于这个VTTTT-anchor primer跟poly-A tail的结合程度
我个人觉得既然你现在做不出来
很有可能是cDNA synthesis这一步就已经效率很低
我当时做RLM-3’RACE的一个原因就是原核没有poly-A tail
没有办法用poly-T的primer来做cDNA synthesis这一步
RLM-3'RACE带来的好处是
mRMA3'连上adaptor以后
后面的cDNA synthesis用的是adaptor specific oligomer
这个效率会有不同
如果已经山穷水尽，这个结果又一定要做出来
可以试试这个RLM-RACE的方法
不过就是多设计个引物，买点T4 RNA ligase而已
Roche的反转录酶还是不错的

另外如果你担心是因为mRNA的二级结构有影响的话
在把RNA加入cDNA synthsis反应之前

这个老板）的文献中对organic synthesis的要求，我也不懂electrochemistry，但我
认为关键要找到二者的结合点。老板要找能够对他的项目有用的人，如果你能够从
synthesis的角度，提供他所需要的而又买不到的只能通过合成得到的新材料，从而深
化他的研究课题的话，老板一定很高兴的。和老板面对面聊天的时候，如果那个老板硬
说，二者没太大关联，你也千万别和他争辩而伤了和气。但化学嘛，总会有些相通的地
方的。
quick learner的展现，只需要一个具体的自身事例。既然是organic synthesis背景，
我觉得很自然你可以从学习操作分析仪器到熟练操作和教导其他team members甚至能够
对小毛病进行初步诊断并维修的角度入手，暗示老板你能很快学习并掌握
electrochemistry的基础技能和仪器使用。这个桥段不要生硬得引入（除非办公室交谈
中老板主动提到），而可以考虑在lab tour走到主要仪器（读老板的paper，找到仪器

major 的人相当，drug design 其实主要是导师做的，但是是我thesis 的重要部分，
我也完全掌握。
听说公司都倾向于招学合成的，然后进了公司在training 他们drug design的知识。要
是把我的project 里面的drug design 部分写到resume和research summary里面，是会
加分还是减分啊？
不知道他们会不会对med chem的人有特别偏见的（因为我知道确实有很多med chem的人
organic synthesis experience比较少）会不会我只要提一点点擅长drug design就给
他们留下这个人其实不怎么会做synthesis 的映象，所以应该只字不提？
搞不清楚，请大家赐教啊 |

1970年代早期，多数中国科学家，在文化大革命中努力生存而无机会开展研究。两位年
轻的研究者屠呦呦和张亭栋，分别在发现抗疟新药青蒿素和揭示砒霜化学成分三氧化二
砷对白血病的治疗作用的过程中起了关键作用。回顾四十年前开始的历程，不乏曲折和
反讽。青蒿素工作源于中国帮助越南抵抗美国，三氧化二砷源于观察、验证和改进乡村
中医的实践。虽然他们的药物挽救了世界上很多生命，两位研究者迄今未获国内外充分
肯定，屠呦呦有争议、张亭栋基本默默无闻。相关的文献埋没于冷僻的杂志和一般不易
看到的内部会议资料。基于原始中文论文、文件和采访，我们在此呈现这些发现的历史
概貌。没有逃脱我们的注意，在古代和近现代中文文献及医疗实践中，可能还有尚待重
新发现的珍宝。
