All the biological process have to abide the law of chemistry.
It does not matter what the origin of a biological process or how it began,
you can say RNA, protein, sugar, or whatever. You get to explain how these
molecules formed in nature in the first place.

中药的科学研究丰碑（修改版）

摘要
1970年代早期，多数中国科学家，在文化大革命中努力生存而无机会开展研究。两位年
轻的研究者屠呦呦和张亭栋，分别在发现抗疟新药青蒿素和揭示砒霜化学成分三氧化二
砷对白血病的治疗作用的过程中起了关键作用。回顾四十年前开始的历程，不乏曲折和
反讽。青蒿素工作源于中国帮助越南抵抗美国，三氧化二砷源于观察、验证和改进乡村
中医的实践。虽然他们的药物挽救了世界上很多生命，两位研究者迄今未获国内外充分
肯定，屠呦呦有争议、张亭栋基本默默无闻。相关的文献埋没于冷僻的杂志和一般不易

际科学界的难题，而这一基础科学问题也与人类、特别是中国人民的健康相关。
过去，世界和中国忽略了1980年代我国科学家在条件艰苦时代做出重要贡献，我们现在
应该赞广西肿瘤防治研究所的严瑞琪和苏建家等、北京第二传染病院的余昌晏等和中国
医学科学院的庞其方等建立乙肝动物模型；
现在，我国已经有条件和能力改革科学管理体制机制、提供适当环境，让一些科研机构
年富力强的科学家心无旁骛，如北京生命科学研究所的李文辉带领学生和同事发现乙肝
病毒受体；
将来，希望全国科研机构能推广适合各个学科和机构自身规律的体制机制改革，使大批
年轻人集中精力在专业上充分发挥作用，他们大有作为才能使中国的科学大有希望。

我用biotin pull down enrich 了一些 ssDNA, 想convert成dsDNA， 然后sequence.
有比较成熟一点的方法吗？
好像很多RNA->cDNA->dsDNA的kit我都不能直接用，因为说first strand synthesis 的
时候，那些kit加了一些特殊的adaptor, 然后在second strand synthesis 的时候就很
方便了。但是我ssDNA是直接从linear PCR 来的。不知道有没有什么好方法做second
strand synthesis。
