自己觉得三项都比较弱，在回复REF的时候也没有大的变化，唯一就是大概引用从９９
涨到１０３。我也看了一些其他网友的经验怎样把自己归到small percentage. 我觉得
太费劲，没有用。我很庆幸自己没有急着回复，让我有机会看到了以下这位网友的经验

但我觉得更主要的进展在于ClojureScript。
Facebook的react.js出来以后，让ClojureScript一下子变得非常有吸引力了，因为
ClojureScript的immutable data structure可以让react.js这种类似3D图形渲染的办
法变得更快。因为在ClojureScript里面，diffing几乎是免费的，只用检查reference
equality, 这就让基于react.js的ClojureScript库比native的react.js还要快，比如
om, reagent等等。
现在纯ClojureScript的库也出来了，甚至比reagent还要快，这样发展下去，用
ClojureScript搞Web UI可能会有大的突破。 |

that I needed for my exp.
however, crosslinking is a tricky thing in that it varies from protein to
protein, and the condition for crosslinking matters a lot too. if you are
doing it in the presence of crude cell lysate, i doubt if there is established
protocols to follow. you probably need to try different crosslinking reagent,
different concentrat |

Write down a short version of your protocol, emit details.
Just say something general like which plasmid to use and what reagents to
use to do what.
Then give him the protocol, and tell him in a very, very nice way:
"This is the protocol for the experiment. I think you should have no problem
with it. And come here, let me show you where the reagents are. blah blah
blah. In case you have any problem in the experiment, you can come and find
me. I will try my best to help you." |

样了。那我在paper里可以这样说明吗？我老板说应该保持前后一致。比如我前面一个
蛋白在无transfection的cells里表达量有较多，但现在在有vector的control里，蛋白
的表达量下降了好多，用RT-PCR检测几乎就要看不到了；老板就要我重做，说这2个
control非得match。大家遇到过这样的情况吗？如果前后不match，该如何说明比较好？

ILMN 的简单 library prep 是多大的竞争优势... 一个不稳定的 workflow
可以轻易毁掉一个几万块的 run, 还赔上几星期的时间。
PacBio RS 目前还没看到有任何人的使用报告。可能都是暗干在心里？ :)
Complete Genomics 的 library prep 会是他们的软肋，还有，他们会有
所有 SOLiD 有的问题, 因为他们也用 oligo ligation.

aliquot出来用。其实做PCR，只要某reagent换了做不出，就肯定是那个reagent的问题
。buffer啊酶啊都不太可能出问题。最容易出问题的就是水和模版。 |

Hi,
I tried a couple of transfections with the NgAgo plasmid from Addgene and
the 5'P oligo against GFP from the paper, but I did not see a dramatic
reduction in GFP+ as seen using Cas9 and a GFP sgRNA.
Anybody tried as well and came to the same (dead) end?

干以后溶在ＤＭＦ 里， 然后滴到phenol/DMAP/DMF里．
或者 直接coupling reagent 用MSNT/Methylimidazole 的组合
弄酯用EDC,DIC或DCC之类的有时候效果是不好，所以 solid phase 上弄酯更多用
symmetric anhydride 或者 coupling reagent 用 MSNT
当然弄成 鲜绿的话肯定最直接，效果应该也很不错，就是有的酸不太好弄成鲜绿 |

安全，所以，在实验室进行研究、完善无可质疑，但如果大面积推广，就应该受到质疑

文章作者的行为和态度相当草率轻浮啊。
很多做法都没有解释原理。有些并不一定对。
任何有危险的工作应该预先详细写下每个步骤。
每个Reagent，纯度和需用量，工具，器皿，手续前后，
时间，温度，防范措施，至少两人一组互相查证。
其它如闹钟或闹表，计时器，灭火或中和剂，等等。
并且预先准备到齐，摆放清楚，适当标明，才开始。
而且在每个阶段，尤其是重要或危险的的，
需要搞个 Checklist。启动前，再三对照，万无一失。
最坑爷的是化学实验室还铺地毯，真是外行的荒唐！
再防火的地毯也无法和水泥或Tile铺面的比安全。 |
