今天收到NIGMS director 的公开信，里面明白着说单个实验室不应该拿到超过三个R01
（除非是clinical research), 并明白指出这个动向的原因是：
1。在钱的总数有限的情况下，一个实验室拿到多的钱就会导致另外一个实验室拿到的
钱变少。
2。大实验室对博士后和研究生培养不利，因为导师的注意力被分散了。
3。从统计上来看，大实验室的成果并不和资金成正比

我还是先抛砖引玉。
半径：
假设车做匀速直线运动，轮子上的力要抵消风的阻力。阻力是一定的。所以轮子上的力
是一样的，但是大半径的轮子需要轴上的力更大（杠杆原理），所以engine rpm 要高一
些（普通na engine）。rpm高了，engine是否efficient要看rpm 的阶段，所以在engin
e最 efficient的rpm之下，是大轮子省油。之上是小轮子省油。考虑到一般efficient
rpm在 55-60 mph，一般开车在65mile以上cruise，所以应该是小轮子省油。对不对？

1.关于standard curve, 我的理解就是确认PCR amplication efficiency,只要你的样
品DNA好的，为什么一定要用实际IP的
样品量（1/20, 1/200, 1/2000 and 1/2000 of ChIP input）standard curve？当然
如果模板浓度太低，amplication curve may be an issue。
我过去的经验是cDNA样品反复冻融会影响amplication efficiency.
此外如果倍数差别很大比如10倍以上的话就没有必要做standard curve，因为即使扩增
效率低也不会影响到最终结果。
我觉得对一对引物做一次就够了。
2 “qPCR本身是有每次
PCR循环的数据的。根据linear range的扩增曲线，是很容易算出primer的efficiency
的（有的软件会自动给出这个值），然后可以根据这个来定量。”
我以前想过机器应该能自动给出这个效率，但从来没见过，楼主能否说出具体的机器或
软件？

Efficiency。这两者基本没有关系。
下载速度跟CPU是否efficient关系不大。cpu再不efficient，它99%的时间还
是在等待network IO。 时下的CPU时钟大致在2GZ左右。即每秒运行400万条指令
（以平均每条指令花费五个clock cycle计算）。
T1 line 是1.5Mb/sec, T3 是44Mb/sec, 最快的t4 是274Mb/sec。
甚至连USB2，也只有480Mb/sec。以每传送一个Byte为一个指令算的话
（实际上每条指令通过Direct memory access (DMA)可以搬运更多
Byte），连USB2都只有CPU速度的1/10。也就是说，你同时连10个USB2
到一单CoreCPU的Computer上，同时从这10个USB2 Drive上“download"
file 到Computer里，看起来这十个Download是在同步进行一样。一个单核
CPU就可以同时handle这么多高速ＩＯ。

第一次做qPCR发现knockdown efficiency不错，细胞再养一段时间后qPCR发现
efficiency变差了。
有人解释说是细胞中KD efficiency高的没有低的分裂速度快，所以养一段时间后KD
efficiency高的细胞就没有低的多，总体efficiency就变差了。
有人遇到过同样情况吗？都是怎么解决的？谢谢！！ |
