发帖数: 287 | 3 有细胞重新做一次么？如果有的话加一个 permeablized cell 做 positive control。
膜破掉了之后 PI 和 Annexin 都可以自由进出细胞，所以这个 condition 应该是
Annexin 和 PI 都 positive。如果这个条件都做不出信号说明你的试剂坏掉了，要是
这个条件有信号但是你的样品没信号那就是诱导凋亡的条件问题。这个实验做完之后不
管结果如何你都有东西和老板讨论了，good luck!

PI+only (%#) --necrosis (dots on the Left side top)(died)
PI+/Annexin-V+ (%#)--later apoptosis (dots on the Right side top)(died)
PI-/Annexin-V+(%#) --early apoptosis (dots on the Right side bottom)(Not died)
PI-/Annexin-V- (%#)--alive & No apoptosis cells (health cells)(dots on the Left side bottom)
I use The sum of "PI+/Annexin-V+ (later apoptosis)" and "PI-/Annexin-V+(
early apoptosis)" to count Total Apoptosis (%)
You do not need do permeablize. If you permeablize, all cells will be red(PI stain), so you can not seperate the lving cells and died cells.
I used Invetrogen or BD kits. You just follow protocol. But I usually use
less quantity as the protocol said. For example, Ann 5uL/sample on protocol,
I used 2.5uL (invetrogen) or 1.35uL (BD)/sample.
PI also is less. 4uL/sample.
They are working very well.
