功的女同事推荐黑豆（black bean),相对高蛋白底碳水。昨天去买菜就顺便买了一些，超
市里还有各种各样的干豆，比较了一下营养表，发现这个red kidney beans 也是高蛋
白底碳水，也买了一些回家。晚上图方便，把black beans 和red kidney beans 干豆
子洗洗就直接扔slow cooker里煮了，想着第二天早上起床就有豆粥早餐多美好啊。结
果第二天吃了大概1/4 cup，吃完没多久就觉得胃胀气难受，头昏脑胀，想吐却又吐不
出来，难受死了。还有轻微腹泻。 隐约记得吃paleo diet的童鞋们讨论过豆子煮不好
会有毒，放狗一搜，果然。大多数生的豆子里含有一个刺激肠胃的毒素
Phytohaemagglutinin (PHA),一种lectin， 豆类中red kidney beans含lectin量很高,
5个生豆子的毒素足以让人不舒服.这个毒素要高温煮才能破坏，用slow cooker低温煮
最糟糕，把毒素都释放出来，比生的豆子更毒。浑浑噩噩过了几个小时后才好一点, 晚

reactor comes with 1.7L pot and specifies boil capacity of 1L.
PCS has 1L pot and 0.5L boil capacity.
I would say it makes a difference if your major purpose is to melt snow in
camp and do not want to spend too much time on pouring water in and out.
1L is also the magic number, the same as a nalgene bottle. If you have a
bottle of water and want to boil it and have it in your cold sleeping bag
before going to sleep, I do not how to do it with PCS but |

以前去过黄石/大倜傥，这次是带父母去。父母岁数大了，所以整个行程安排比较松快
，也没有hiking的内容。住lodge/hotel，带oven/电饭煲做饭，也在外面吃吃。
大约提前4-5周开始预定机票和公园里的lodge。机票比较贵，旅馆相对比较便宜，8个
晚上，平均$100。在SLC住一晚，黄石里Lake Lodge 3晚，Canyon Lodge 1晚，Old
Faithful inn 2晚，Jackson hole一晚。
租车是在版上取的经，full size car, 9天，$277 (alamo),提前10天左右订最便宜。
汽油的价格在$3.43-$3.69/Gal之间，在盐湖城，Jackson hole, wy和Gardiner, MT加
的油，共用了$180的汽油。

怎么个crosslink法？我是加了1st Ab overnight再加beads的。
还有elute我是用的SDS sample buffer，然后就boil，100 C，10min。boiling beads
和elute是两个独立的过程呀，为啥elute可以取代boiling？是我理解错你的意思了吗？
关于ECL plus，我觉得用ECL曝光久一点，貌似也跟plus一样的效果，而且plus还会令
blot颜色深浅不均一，不知道你会有这个问题吗？如果有，怎么解决比较好？

对3x的etn/etf来说，主要有两种损耗，contango引起的，和震荡损耗。
contango不利于ugaz，但有利于dgaz。不过长期来说，震荡损耗应该是大头。dgaz的震
荡损耗比ugaz的大不少（理论上两倍）。
搞一点理想化的数学计算吧。比如开始气价是3, 先跌到2.5，再回到3. 假定大跌在一
天完成，大涨在一天完成，那么震荡损耗

上次一样又是完败，这个季节和100多度的高温，striper bass boils 已不可能
，约好明年春季一定回来报仇。
完败是对 striper bass说的，其他鱼可是上了不少。我的收获是见识到utah钓鱼牛人风采。我在鱼版常贴的trout，小bass 什么的根本不入他们法眼。看了他们往年striper bass boils 时钓到的上百条striper bass 还有硕大的walleye的照片，才知道在 utah 钓鱼原来要这么玩才过瘾。
我们一晚上上了无数catfish，他们自然一条都没keep，每上来一条先问我要不要keep，我不要的话就扔回水里，不记得release了多少。因为我是第一次钓到catfish，还是兴高采烈抱回家12条。我的其他空白是第一次上LM bass 和第一次上传说中的 tiger trout。世外高人是不会在这里贴照片的，我作为新手当然还是要一往如故的BSO一把，看图吧。 |

发帖数: 2352 | 35 上周末出发去的黄石，走之前做了一下功课，刷到了四天园内的旅馆，在不同的口，预
计从东口进，到的时候是晚上，后来朋友说东口晚上很难开，于是修改路线走南口，同
时把第一天在Lake Lodge的旅馆取了，换成南口的Grand Village，一路还算顺利，到
的时候已经晚上九点多了，屋子里没有空调，但是配了一个风扇，有暖气，在湖边的那
个Lake House打发了晚饭，有Trout，发现鱼肉块很大，比俺平时钓的大的多，问了一
下才知道是Lake Trout，品种不一样，是黄石的特产.
早上大概7点多起床，到附近的Visit Center去看了一下，有工作人员讲解在园里的安
全问题，说熊的速度要比百米冠军快好多，遇到了只能自求多福了，倒是在拿材料的地
方看到了墙上的熊皮，讲解员说是真的哈.
先到的Old faithful,到了没多久就是喷发，人很多，喷的不太久，也没有图片上喷的
高，之后就按照网上的帖子，走边上的Trial，来回一共用了三个小时，回来才知道大
概单程有2.6miles，女儿最后都不想走了，路上的Daisy Geyser喷发的时间倒是很长，

推理小說萬流歸宗，一切都由福爾摩斯（Sherlock Holmes）而來，讓我再從福爾摩斯
講起吧。
在塑造神探福爾摩斯這個角色時，類型開山祖師的英國作家柯南．道爾（Arthur Conan
Doyle, 1859-1930）不僅要他獨特，讓讀者一讀難忘，又努力想要讓福爾摩斯角色飽
滿，讀來可信。為了讓福爾摩斯獨特，這似乎不難，光憑他看一下你的袖口就能說出你
的職業，這種本事還不算獨特嗎？但是這樣的「新人類」要怎樣讓他可信？你要我怎麼
樣相信世間真有這樣的血肉之軀，行走在你我之間？
柯南．道爾其中一個重要的努力，出現在史上第一本福爾摩斯小說《暗紅色研究》（A
Study In Scarlet, 1886）裡，那就是原作的第二章裡提到的「福爾摩斯：他的界限」
（Sherlock Holmes: His Limits）。
所謂的「福爾摩斯的界限」，指的是福爾摩斯的「知識邊界」或「知識範圍」，那是福

不做统计拍着脑门估计一下，我们吃得东西可能有一半都是煮着吃的 — 从早上稀松着
睡眼煮碗麦片，到忙的焦头烂额煮包方便面；从到好友相聚热火朝天煮个火锅，到烧包
的难受去点一个佛跳墙； 从夏天清热祛火煮个绿豆粥，到每日电饭锅煮来香香的白米
饭……可单一可混合，可简单可复杂。煮恐怕是最常见的烹调方法了。
煮之所以可以大行其道，很大程度是因为作为技术它比较简单，把食材和水放在容器里
加热一段时间就行了。小时候我开始进厨房做吃的，就是从白水煮蛋开始的。但若说煮
是最简单直接的烹调方法，却又不太对了。 在人类饮食历史上，煮出现的时间要远远
晚于烤 - 若想煮东西当然先要有容器，制作不漏水而且耐加热的容器，只有当人类文
明发展到相当的高度才能出现，所以别看日后煮饭几乎成了做饭的代名词，其实我们开
始好好煮东西吃其实也只有几万年而已。
有人类学和考古学考证说，其实在人类可以制作容器前就已经会煮了，但是多要依赖于

I have a similar one that won't take that long:
1 hen, put in a pot covered with water, cook to boil, get rid of water.
Change water to cover and let it sit at room temperature for about 30
minutes. Get rid of water.
Add new water to cover the hen, add some green onion, a couple pieces of
ginger root. Bring it to boil, turn the heat down to let it boil for another

有GFP, Flag等tag.Overexpression 做Co-IP.
先做A和B的相互作用，用protien A beads，IP后boil.先IP B, WB A.对照有（1）non-
transfected, （2）only transfected with A, 试验组（3）transfected with A and
B. IP后发现（1）没有A,（2）中有weak band of A,（3）中有strong band of A.
然后怀疑A蛋白太大，可能和beads或者antibody for IP有non-specific结合，所以（2
）中有weak band of A.之后change strategy: IP A, WB B.后来发现有同样情况。
请问下大家有没有发现蛋白和protein A beads non-specific结合的情况？

谎言之九：普兹泰的小白鼠实验结论不成立
【普兹太在电视上宣布他的实验结论的时候，他实际上还没有完成全部实验。按照
科学界的惯例，他应该在完成实验之后，写成论文，经过同行审稿通过，在学术刊物上
发表论文，然后才向大众媒体宣布他的发现。普兹太所在的研究所的领导见他违背学术
规范，向公众提前公布未成熟的实验结果，引起不必要的恐慌，觉得他败坏了研究所的
名声，决定给他处以停职的处罚，后来又强迫他退休。普兹太当时已经68岁，本来也该
