We use same inducer for all the experiments.
P should have same function with respect to X1-R2 and X2-R2. Once bound with
X1 or X2, both R1 and R2 should block the transcription of eGFP.
R1 and R2 recognize different inducer. The inducer for R2 is a universial
signaling molecule within all living organism.
One model currently I came up with is that X2 will interact with X1 and
decrease the binding affinity between R1 and X1, thus we saw an activation

Mg-binding UV不会有变化，intrinsic fluorescence值得一试。或者用能bind Mg的
fluoresent probe做competition titration.又或者equilibrium dialysis.ITC,NMR也
可以，但是不容易做，蛋白需要量也大。
还有一个值得一试的是：用Co先代替Mg作titration.Co binding通常伴随UV变化，然后
加Mg来compete with Co.这样数据分析也能得到Co/Mg binding constant. |

了和PPARg binding motif overlap。
文章找到一个tissue specific transcription factor。KO和overexpression都有
phenotype，phenotype还是和fat development相关的，临床意义很强。还做了chip－
seq发现bind tissue specific genes 还和已知的 master regulator binding region
重合。很strong了。太多的CNS有个比较强phenotype却没什么explanation的。
一座的还是个中国人。不知道会不会上网和你argue一下。这个放在Cell或者Nature中
，质量也是上等的。 |

从你提供的信息看，这个磷酸化有两个作用：
（1）磷酸化->稳定蛋白—>DNA binding增加，那么DNA binding增加就是由于蛋白多了
导致的（相当于overexpression）
（2）DNA binding 之后，转录活性降低，说明磷酸化抑制了transactivation
(1)和（2）不矛盾，结论应该是磷酸化的overall effect还是抑制转录活性 |

我也是最近分析自己的RNA-seq数据，想到TFBS这个问题。
我得到很多基因可能是被wnt/beta-catenin激活的。beta-catenin不是直接的TF，只是
co-factor，结合了TCF/LEF之后才激活下游基因表达。所以接下来很自然的发现，在随
机检查过的十多个基因第一个exon的上游，有很多predicted TCF binding site。我看
了上游3K的样子，没有海胆的TFBS预测软件，只好纯人工找。。。
然后我就在想那么control是什么？那些下调的或者其它细胞里没有被上调的基因？结
果我又手动检查了十个左右基因，我了个去，同样也有类似水平的TCF binding site。
。。后来老板提醒才想起来，TCF结合了起到抑制作用，后来beta-catenin结合上去了
才会有转录。所以真的就像你说的，binding不等于对基因有直接的调控啊！
这样看来，像我们海胆领域，每做一个TF的knockdown，overexpression都要做一大堆
in situ来看哪些基因真正受到调控，虽然费时费力，但是真的是最直接的证据。这点

马英九指出，这个法庭的存在，应该是为国际争端定纷息争、避免走上冲突甚至战争。但是，这样的仲裁结果，不但没有排难解纷，反而增加了冲突的可能。「在背後操控的势力，看似赢了面子，其实却输了裡子。」
「菲律宾对中国」南海仲裁案，常设仲裁法院今天公布结果，称中国大陆在9段线范围内主张的历史权利没有法律依据，而南沙群岛的所有海上地物，均为礁岩，包括太平岛在内。
马英九晚间在脸书发文表示，从去年12月到今年5月，包括他在内，150多名部会首长、国内外学者专家和国内外媒体，都访问太平岛，亲眼见证这个淡水充裕（南沙唯一）、农产丰富、生活机能完整的岛屿，完全符合「联合国海洋法公约」第121条关于「岛屿」必须「维持人类生存与自身经济生活」的要件。
他说，至今年4月底为止，也有430篇国际媒体完整陈述「太平岛是岛」的事实。太平岛是「岛」不是「礁」，已是国内外多数人的共识。

经过一个中间宿体性的其它动物才能传染给人。所以，病毒发生后，就故意误导全国人
民去海鲜市场找证据，找其它野生动物的麻烦。 而且还是病毒所去找的，找完了还装
模做样化验呀，分离呀什么的。最后把责任全部推给了海鲜市场的动物。可是那种动物
，一直不敢说，说了其它相关已经在人就会去早那种动物监测。 所以压根不说，打马
虎眼。
事实上，早在2013 年，就是这个武汉病毒研究所，已经从来自云南的蝙蝠身上所携带
的corona virus中分离出第一株蝙蝠SARS类似样的冠状病毒的活病毒，其中就包含了类
似于S类型的基因。从而证实这株病毒能够使其接受和SARS病毒相同的受体，并能够感
染人的细胞。对此新发现，武汉病毒所还把它以武汉病毒研究所的英文简称命名“WIV1
”，以彰显这一发现的重要价值和属于自己第一个发现的巨大研究成果。这个成果刊载
于2013年11月的《自然》杂志。

What I want to know is, is this ruling binding ONLY on California, or is it
binding on all the states within the ninth circuit? Are they saying that
Proposition 8, itself, is unconstitutional, or are they saying that all bans
on same-sex marriage are unconstitutional?
As it is, it seems they're only saying that this particular ban was
unconstitutional, since its proponents failed to offer compelling evidence
