overexpression 不是确定tumor suppressor的好办法.比如,有的tumor suppressors
overexpression 就没有 phenotype.
你可以先siRNA knock down 这个基因,看看细胞增殖有没有增加,如常用的BrdU
labling之类.
此外说不定这个基因tissue specific的tumor suppressor,不一定在所有细胞系里面都
有作用. |

有 overexpression来研究的。但我的二老板就是认为是我本身的原因做不出来，对我

但我們實驗室的concept 就是， 既然叫 tumor suppressor 基因， 就應該有suppress
的function, 所以就一定能做出phenotype.
這個基因當然有可能我們挑的不是一個 right 基因。 但因為整個缺失的region 只有
五個基因，只有這個基因看起來比較象。可能性還是比較大的。
我还想請教各位，如果你們實驗室發現某染色體缺失或擴增跟tumor 相關，通常是怎麼
著手去找drive gene的？ |

其實我已經在準備走人了。只是覺得這個project花了很大勁做成這樣，心裡不大爽，
还想再試一把，另外也覺得如果能出點東西再走不致於讓老板很生氣。
本來想來這個實驗室學點 cancer stem cell 的東西，希望以後回國能用來忽悠人。結
果比較失望。
我其實每天都在努力看很多paper, 然後跟老板們討論。悲哀的是他們根本不看paper，
並且沒有經驗， 沒辦法討論。 我以前的老板非常聰明，新老板，接觸的東西新而多，
我只要跟他一提，他馬上就明白， 我現在這個大老板，我有一次看到cell 上一篇
paper 很有啓發，很興奮地跟他們討論，大老板聽了半天，甚麼也沒明白（他連
transfection 是啥也不大明白）， 最後还發了一通脾氣說我英語不好。
我跟我以前的老板在科研思維上可能比較接近，所以總能達成共識，效率很高，以前的
几年基本一年一篇不錯的paper。但現在這個二老板的，我覺得他的建議我總是覺得很
別扭，但不做又不行，所以就不想聴他的建議了。比如說他覺得這個基因在肺癌細胞表

2mm suppressor压力会太大，2mm柱子配4mm suppressor也会back pressure不够影响分

close-medium range p90+holo+suppressor. i don't use tac light or laser at
all. they give my position away too easily. same gun for engineer or assault
class, depending on the need. i have a soft spot for p90 from CS days.
long range i use assault w/ an94+4x+suppressor+grip+m320. since i play HC
only, the 2shot burst kills. very deadly. repeated burst is very fast with
this gun too if you run into someone around the corner.

如果已经抑制tumor growth了，甚至导致tumor shrink了，tumor cell数目已经减少了
，那可转移的tumor cell也少了，不是也会导致metastasis减少了吗？
我做的这个基因是tumor suppressor and/or metastasis suppressor, 这个才是我做
的方向, 有好多老鼠的活要做.
embryonic lethal这种事有点难, 主要是我没啥development的经验. 先干着吧. 我本
来跟老板说我不想lead这个project, 现在估计只有我能lead, 除非老板再招一个人干
活, 我就可以把这个脱手了.
上个月刚刚写了一个新的关于cancer的animal protocol(实验室以前没有独立的animal
protocol), 还好以前phd的时候写过不少类似的. 现在又写一个development的

不过总体来说, metastasis suppressor 比较难确认, 因为in vivo的实验是必须的,

这也是一个证据表明，有其他不依赖端粒长度的衰老机制。
因为人的端粒长度很短，随着年龄的增长，端粒不断缩短，尤其是在快速分化增生的器

当然tumor suppressor 的激活是双刃剑，一方面抑制肿瘤发生，一方面导致衰老。
所以，如何正确激活干细胞的tumor suppressor，或者如何fine tune tumor
suppressor 将是一个非常有意思的研究衰老的方向。

(red) tubing and the smaller (2mm) suppressor as opposed to the macrobore (
black) tubing and the larger diameter suppressor (4 mm).我们现在用的是ICS-
1000的Ion Chromatography,需要把apatite里的calcium, ammonium and sodium分离出
来。我已经给dionex的technician service 打了很多次电话，他们都说啥样的
suppressor 配啥样的tubing就行，除了flow rate不一样，其他的好像没什么区别，但
是老板还是觉得肯定有advantage和disadvantage，想找出我们到底应该用2mm的
suppressor还是4mm的？
有没有人可以给点建议？谢谢！ |
