critical and is way less appreciated by the cancer research community. In
my own experience, loss of some tumor suppressor protein function cause
less proliferation but also cause dedifferentiation.
proliferation和apoptosis被“重视" is because the phenotype is easily and
quickly characterized. But cancer is the a chronic disease so slow
proliferation does not matter at the initiation stage. |

化。做了免疫能力测试，footpad assay，发现ko小鼠免疫能力下降。 老板想让我去测
免疫相关基因western，我测了nfkb，没有变化。
请问还有什么其他重要的免疫系统基因吗？

“北京协和一个毕业生因为两篇论文造假被波士顿大学剥夺教师职位”。
王盛毕业于北京协和医学院，获得博士学位，到美国后在哥伦比亚大学和纽约大学做过
博士后，2000年受聘到波士顿大学医学院癌症研究中心工作。他在乳腺癌研究方面有不
少成果。
王盛涉嫌造假的这两篇论文，第一作者均是张葆华（Baohua Zhang，音译）。张葆华是
王盛的妻子，1984年北京医学院牙科专业毕业，其后在北京协和医学院获得硕士学位。
不但是“协和有人造假”，而且居然是“协和毕业的一对夫妻合伙造假”，奇闻也！
现在韩春雨和沈啸两个作者也是协和人，成双出没是协和人造假的特征？需要N+1的例
子。

发帖数: 1 | 20 实际上我们对cancer甚至整个信号传导的理解还远远不够。我们的知识是碎片化和模糊
的（等我们真正理解之后甚至会发现是错的）。比如一个简单的问题，像Myc, Rb和p53
在基因调控网络中的真正角色是什么我们并不清楚，这不是简简单单一个术语，像
oncogene或者tumor suppressor能概括的。就像组装一个飞机，手下有一大堆零件，但
是只有它们是正调控或者负调控原件的信息，这个飞机拼起来之后肯定要摔下来。
解决这个问题， 一个就是要用更精细的测量方法，和更基于物理事实（有关生物的就
是热动力学，化学反应动力学和统计力学了）的精确数学描述来定量化研究这些问题。
统计学其实是数学中的麦丹劳，是用来寻找相关性的。而只有基于物理定律的数学表达
才有预测的能力。可惜大部分学生物的人数学训练都不够。

尽管科研的目的是要尽可能接近的呈现事实。但是不同的reasoning， 虽然可能都是
causal的，但是逼近程度可能差的很远，因为reasoning是以concept为基础的。
这么说吧，对现象的任何一种解释总会用到一些术语，但是你仔细推敲起来，并不是每
个术语都靠谱的，有的是自然地，有的则是人为创造的并不一定靠谱。比如我跟你说基
因p53，这个描述是准确的，因为你知道它对应着一段DNA序列，是一个特异的东西。而
我要跟你说oncogene，则相对p53不那么靠谱，因为这个术语是根据phenotype定义的，
这一类基因activate之后会促瘤，对应的还有tumor suppressor。可是事情又没有那么
简单，有些基因在一种情况下促瘤，在另外一些情况下又抑制瘤，那你该怎么定义他们
呢。可是如果你换一个paradigm，在系统的角度下看待这个问题，它的角色或许完全就
由它在网络中所处的位置决定，它的不同角色只是系统运行到不同状态带来的结果，这
或许就能unify了，而且听起来也很自然。

还有一种情况就是 gestational mutation，胎儿发育过程中，正好在致癌基因上发生
突变（oncogene)，孩子从生命一开始就完成了多数癌症病人到中年才开始的癌变进程。
人体当然有清除癌细胞的机制。防止癌变是多细胞生物的一个重要任务，有非常多关口
来把关保持genetic stability. 当恶性肿瘤发生了，就说明这些关口一道道都被突破
了。这个癌细胞已经是“羽化成仙”了，在这个宿主体内实现了immortality(永生）。
化疗，手术，放疗一切目的都是为了尽可能清除这一株细胞。留下几个，都有可能会卷
土重来。
这是为什么癌症是绝症。
寄希望重建平衡是幼稚的，癌症发生的过程就是一道道攻破防御机制的过程。che

在做相关项目，上来胡说几句，当是抛砖引玉，大家多多指正
otto warburg老早就发现cancer更喜欢glycolysis，还因此得了诺奖，之前的肿瘤代谢
主要在体外造影上有一定的应用，为啥肿瘤代谢要到现在又火了呢。在这个field里真
正让代谢又火起来的个人看来是三个topic: PKM2, IDH1/2, Lkb1/mTor/AMPK。
尤其是Lewis Canteley 2008 Pkm2的nautre，让人们知道了cancer cell是如何建立
Warbug
effect的，这之后各种可能和warburg effect相关的pathway又被人们陆续挖掘了出来
