要东西，老板本人也不出面，postdoc整天发信。我老板答应了给老鼠，cc给了我。结
果他们说不要老鼠，要把organ 取出来，mesh好了，把细胞寄给他，他等着revision。
我心好也就答应了。结果然后他还要Immunization 的reagent。昨天刚给寄了 ，今天
又发信，还要detect的reagent。这些reagent都不是commerical available的，都是自
己conjugate的，还要过colomn，做QC，折腾了很久才搞到一点点。 |

neurons. None of them gave me a decent efficiency. NeuroPORTER (Genlantis or
Sigma-Aldrich) was the best (~30% in SH-SY5Y, 18% in Neurons using FACS).

I worked for bio startup, so I know a bit. There are many regulations. Let's
not even talk about how to get your reagent. Assume you can ship to your
friend's lab, and then bring to your basement. How are you doing to dispose

plate, 每一孔我用了4 ul, 好像再多的话细胞很快就死了， 然后siRNA 从10 nM 到
100 nM 都式过。 我的问题是， transfection reagent 和 siRNA之间是不是应该有一
个固定的比例呢？ 比如 10 nM siRNA用 4 uL transfection reagent, 20 nM 用 8 uL

Ramp rate (C/sec) 20 2
PCR volume 10 ul 10 ul
Amplification success fail
The reason we changed the ramp rate is that 2C/sec is recommended for the

acid (TFA)的溶解能力特别强，很好用。可是，高分子（白色粉末）溶解于新鲜的TFA
之后溶液由无色透明慢慢变为淡黄色透明。
是不是因为TFA是强酸有所谓的发烟现象？浓盐酸就是淡黄色，因为含有少量杂质Cl2
。另外， 含卤素或含硝基的化合物容易变颜色,是少量变质导致的吗？

病的高危人群。可能得的原因中我占了三。。具体的就不说了。） 还有3周做糖筛，我
想现在就买个血糖仪或是血糖试纸， 想看看要是血糖高了，就严格控糖了。 其实就算
是不高，我也应该控制了。（自己估计血糖低不了。）
用普通的尿液分析试纸能不能大体看出血糖和尿汞？在OB那里看到他们每次测尿也就是

刚加入这个新实验室一周，每天去上班都有点硬着头皮去的感觉，感觉这个实验室的风
格比较难容入。
我是换了一点方向的，所以新东西会比较多；这个实验室的人和老板很喜欢问一些问题
：比
如why is that? what is this reagent? what does this reagent do?
读完老板给的paper后，老板立刻会上来和你聊，而且还会问很多figure和方法的detai
ls，感觉就像Phd preliminary test一样。有的问题一时不知道怎么回答，老板有点咄
咄逼人，让人下不来台。 |

发帖数: 741 | 49 我说两句。我原来是在Biotech，管过Engineering和Instrumentation。不客气的说，
可能你没有实干经验，至少是商业化的。
假设你配简单的试剂，比别人的都好使，大家都喜欢。问题：你有生产条件吗？别告诉
我你就在自己的实验室里瓶瓶罐罐。你怎么保证没有Contaimination？至少要有
Asceptic filling。
如果有Bio Reagent，或者Chemical Reagent,运输，储藏，很多都是需要Register，或
者License的。
上面是最好的假设，是假设的你东西真的有市场，而且用户愿意接受并更换。但是，这
个是Big If。事实上，所有公司卖试剂，都是从捆绑在仪器上开始的。就像打印机靠墨
盒赚钱那样。
这些看法，不是想打击你。很可能你的试剂几年后大卖。 |

自己觉得三项都比较弱，在回复REF的时候也没有大的变化，唯一就是大概引用从９９
涨到１０３。我也看了一些其他网友的经验怎样把自己归到small percentage. 我觉得
太费劲，没有用。我很庆幸自己没有急着回复，让我有机会看到了以下这位网友的经验
铁，受益匪浅。
