上限。如果不promote她，她以后的加薪都只能以lump sum的形式拿，而不是计入薪水
，对她很不公平。
如果promote她，别的小孩可能不高兴。因为这个人工作态度虽然努力，能力上并没有
特别突出的地方，整天就知道说，到底我要怎么做才能升职，你给我个单子我就照着做
啊。而且因为过考试快，她已经比其他人赚的多多了。她的reputation是喜欢跟别人比
，事事都要拔尖，而能力一般。如果promote她，一是不公平，而且我担心会让大家觉
得，会哭的孩子有奶吃，然后全都跑来哭。
另外HR不同意调整薪水上限。这个已经试过了。

上次是点进去live chat，然后把账号的资料告诉他们，询问为何收费会高了这么多（
其实也就是扯淡，本来就知道在promotion过后价格就翻倍了）。在知道了原因之后，
我问有没有什么方法可以降低点费用，然后它就给出了一个三个月的promotion加速度
，接着我就接受了。它们就在网上把我的账户改好了。
最近一次是帮朋友，两个人一起去的办公室，就跟去菜场买菜一样问能不能便宜点，那
个亲切的阿姨也给了一个三个月的promotion。
不过我用的是只有网络，平常就一网虫。不知道bundle的有没有什么区别。
至于换账号之说，是当时开户的时候那个来装的人说的，此事还没有具体去实施过，嫌
麻烦。不过应该要去面谈，至于要不要还modem再装一个就不清楚了。
有一次在网上乱七八糟的弄了下，结果有个modem寄来了，还回去了…… |

Promoter序列的结合，不明白人们为什么倾向于COS细胞而不是别的细胞例如HEK293?

以前有人做过比较小的promoter效果不好 于是乎就不断增加promoter的长度
似乎已经n多Kb了还是不够specific
个人感觉虽然长度增加了但很多都是没用的序列 关键的elements还是有可能不在里面

做reporter。阳性的对照用的是CMV作为promoter，能看到GFP，但是效率非常。查了文
献，有的说CMV能在S2里表达（但量比较低） 有的说不能（那里说的是不能再KC细胞，
没有专门说S2）。所以想问下 有做过的 给个准确说法呢。 另外我要是就只想看看
有没有promoter activity的话，只用GFP作为reporter 而不是GFP跟其他蛋白的fusion
做reporter 是不是也可以的呢？ 新手呢，求狂拍 |

），这样不会引入多余序列。
如果有得话可以用bsaI方法构建包含你promoter的片段，然后用各种方法和其他部分连
起来
另外promoter不长得话也可以直接合出来然后用PCR和原来部分连起来（有些时候不太
容易，promoter 或者重复序列repeat次数超过需要，不过一般换换条件啥的总能挑到
