做phenol extraction，氯仿这一步是不能省的，否则除不干净phenol。
乙醇蒸馏是和水共蒸馏的，最纯只能达到95%。工业上制备100%纯酒精是要在95%的基础
上再除水的，我忘了是怎么除了，反正依稀记得说是最后出来的纯酒精里，会有痕量的
苯。一般如果用做溶剂没什么问题，可是如果用来做DNA沉淀的话，如果酒精的质量不
过关，反而会影响后续的实验。 |

这个星期第一次做CHIP.完整的protocol就不贴了，但：
1. reverse crosslink 在65℃上热了16个小时。
2. 然后加proteinaseK在37℃热了2个小时。
3. 纯化DNA用的是phenol/chlorofoam.做这一步的时候有点怪。加了phenol/
chlorofoam然后spin down，可以看到aqueous layer和organic layer,但中间的那层
lipid layer基本上就没有。
最奇怪的是phenol/chlorofoam的最后几步。我把aqueous layer提出后加ethanol+NaCl
+glycogen, 然后放零下80℃三个小时后spin down, 居然看到一个有我半个小拇指指甲
大的白色的precipitate.当时我就觉得有点不对劲。但我用70%ethanol洗了一遍之后，
这个precipitate的体积马上减少了90%多，颜色也变蓝了（我们用的glycogen是蓝色的
glycoblue).之后我再洗了一遍。

俺硕士还是肄业，怎么懂得如此高深的东东。。。
不过放了半天狗，查了半天金镐头数据库。。。
超工发现：
这个体系不能用DMAc啊。。。
1. DMAc稳定phenolate啊。。solvation shell大啊。。反应就慢啊。。。
2. DMAc稳定反应中间体啊。。不容易进行消去Cl或者phenolate
3. phenolate也是好的离去集团啊。。
当年俺用一个malonate的酚酯的底物，加碱马上消去形成ketene了。。
综上所述啊，不反应不奇怪啊。。。 |

我觉得比较重要的是第一步trizol
（或者别的phenol based reagent不管每个公司起的什么名字）
和起始材料之间的比例比较重要
另外我自己做过对比
phenol based方法不走colum比走column产量多出来至少5倍以上
对于起始材料足够多，或者下游实验对于产量没有太大要求的实验来说自然column方便
省事
不过有时候还是需要用old fashioned method啊 |

Cartilage contains a lot of proteoglycans and glycosaminoglycans (GAGs). I
first homogenized cartilage into powders by freeze-milling. I then tried
guanidine and phenol to dissolve the cartilage powder but it was not quite
efficient. Most of the 100mg of cartilage powder was not dissolved in 2 ml
guanidine/phenol (Qiagen tissue lysis reagent).

Input DNA，从来不测ChIPed DNA
Nanodrop测得是OD吸收值，用phenol的话，会有一些phenol或者没取干净的蛋白质类的
也会吸光，影响浓度。
对于ChIPed DNA，我们这的core facillity用Q-bit，Q-bit更准，因为据说是根据与
DNA特异性结合的原理而测的，具体是啥我忘了。
我这有个例子你可以参考，上个月我的数据，
测ChIPed DNA浓度：nanodrop：15.3ng/ul, Q-bit: 0.236ng/ul
对于看不见沉淀，你加carrier了么？glycogen or yeast tRNA之类的。 |

污染：
一半废水进入地下水；地上浓缩的废水敞口蒸发；大量天然气和其他有毒气体直接泄进大气。

审查：cheney在任上给这项技术立法大开绿灯，使其对任何环保法案免疫，同时不需要公开使用的化学成分。EPA更是连审查都管不着。直到2010年6月hydraulic fracturing使用的化学成分才公开，其中相当多的都是致癌物，绝育变异，不一而足。

台塑於2008年6月取得越南政府許可，在越南河靜設置鋼鐵廠，是台塑集團首次跨足鋼
鐵業，也是越南最大的外資投資案，不料在2016年4月，台塑被發現排放污水到附近海
域，造成越南中部沿海2百公里汙染，因此台塑被越南政府要求支付5億美元（合新台幣
約161億元）補償金(連結)。
汙染影響沿線兩百多公里，受害地區包括義安、河靜、廣志、廣平、順化等省份，眾多
魚群暴斃，影響數十萬居民生計，據2016年5月統計，死亡的漁產超過100公噸（連結）
；越南政府總理阮春福當時表示，這是越南有史以來最大的環境公害事件（連結），被
稱為「福爾摩沙災難」（Formosa Disaster），又稱為「台塑災難」（台塑企業，

frozen Herring from China under National Security. But China doesn't export
any herring of any kind.
