- mycoplasma: 把最新1月份冻下的vial送去检测，没问题；昨天把刚刚在culture里死
掉的细胞又送去了，还没拿到结果；
- 液氮:应该不会，实验室的液氮罐还是维护的不错的；
- 我的具体cell line是Flp-In™ T-REx™ 293, invitrogen的，非常容易
脱离plate，还很敏感：density太低会不爽；incubator水不足了会马上不爽；拿到
incubator超过15分钟也会不爽。总感觉这293没有传说中那么容易操作。。。也许最基
本的方面就有问题？ |

Hi,
I tried a couple of transfections with the NgAgo plasmid from Addgene and
the 5'P oligo against GFP from the paper, but I did not see a dramatic
reduction in GFP+ as seen using Cas9 and a GFP sgRNA.
Anybody tried as well and came to the same (dead) end?

293T cells are maintained in high-glucose DMEM (HyClone) supplemented with
10% FBS (HyClone) and penicillin/streptomycin at 37°C with 5% CO2
incubation. Cells are seeded to 24-well plate (Costar) with 60% confluence 8
hours before transfection. Thirty minutes before
transfection, cells are washed twice with PBS and medium is changed to high-
glucose DMEM medium containing 2% FBS.

今天重新做了1-2小时的fpg incubation，最后得到了很好的预期的结果，就是（fpg -
mock）和 （fpg+ 样本）在qPCR后Ct有明显的3-4 cycle的差别。
然后把这些genomic DNA都跑了1%的gel ,如图。lane1（mock）和lane2（+fpg）是切一
小时的结果；lane3和lane4明显什么都没有，lane5是positive ctrl。
还是很惊讶nebuffer+BSA incubate genomic DNA overnight会导致DNA degradation；
另外如图里lane1和lane5都是标准的gDNA gel pattern就是有个23kb的sharp bands，
而lane2中因为fpg treat所以high-molecular-weight band都被切没了。不知道这解释
对不对 |

提到的潜在买家中，有一家叫global foundries最有可能中标。大家如果开车路过237
和great america parkway，会注意到那里在建一片新的玻璃幕墙的大楼，差不多快建
好了。其中有一座上面巨大的logo，就是global foundries。他们原来的办公楼在237
上880的地方。实际上，就在几天前，我在多个贴里连续提到过这家公司，因为其最近
新请了一个ceo，就是之前摩托罗拉移动部门卖给google时的ceo。一个确实算是精英的
老印，在高通从一个底层工程师迅速做到vp级的高管，后来被聘为摩托罗拉移动公司的
ceo，再后来把moto卖给了谷歌。

Last year, a group of Media Lab students visited Shenzhen with, bunnie, an
old friend and my hardware guru. He's probably best known for hacking the
Xbox, the chumby, an open source networked hardware appliance, and for

第十八届执政集团登台亮相，李克强主政的中国经济开始变化思路和频道，开启了改革
开放以来最为艰难的经济改革。也是在这一背景下，近两年来，中央出台并加大力度推
进“大众创业、万众创新”活动，并上升为特定时期的国家战略举措之一。中央此举的
重大意图体现在四点：
一是为体制外新生的经济体，提供全方位、大力度的扶持，用创新增量“弥补”传统存
量的萎缩；
二是在旧的经济体制不灵而又无能为力的情况下，引导各级财力及资源配置向新的经济

众多顶尖名校里学术积淀是最少的，整一个暴发户似的，也就是过去三十年才渐渐超越
Berkeley。它一直靠硅谷的资金吸引各种人才，但学校本身的氛围浮躁的不行。去ee和
cs系看看就知道了，学生一天到晚意淫开公司发大财，你们眼看的少数几个公司成功了
，也有大把毕业生搞了些垃圾startup还美其名曰高科技。斯坦福其实跟USC很像，都不
是做学术的地方，最多是一个大型incubator。 |

目前喜欢当教授的多是农村娃，自己父母辈是城市里面当教授的，小孩也常常不愿重蹈
覆辙。只说大陆华人，不讨论台湾人和美国人，这些人已经整体富裕了好几代，部分人
慢慢回到学术界正常。
这是一个观察结果，拿来跟大家讨论。本想发在fuckty版逗逗那些自我感觉良好的小老
师们，不过又烦那几个跳梁小丑纠缠，什么我家不是农村的是县城的。蒙脸好说话。
F版的帖子经常上首页，进去就会发现一帮傻叉上蹿下跳，一副老子天上事情知道一半
，地下事情全知道的样子，直欲干呕。偶尔有个言语风骚的大娘就捧上天，一些乐于照
顾成全学生的老师偶尔说几句不同意的话就要被踩到地里。唯我独尊的态势怎么看怎么
眼熟，今天我突然顿悟，这泥马不就是凤凰男/女的嘴脸么。逻辑分析一下为什么农村
娃喜欢当老师，先说清楚这些是大势，不用来争什么个例：
