incubation 过夜70度,发现tissue结构收缩,如附件所示,图示是我已经完成整个in
situ的结果,事实上我在incubation以后就已经发现tissue结构收缩了.
我问了一些人说需要在fixation的时候做decalcification,我做了decalcification,结
果in situ 的时候tissue还是会收缩,事实上看起来更严重.
我不知道这究竟问题出在什么地方,看起来像是tissue attach不牢固,导致容易变形.请
问有什么方法可以使tissue attach牢固么?
我是根据下面的步骤做的:
1. cut the arms and legs
2. 4% PFA (pH 7.1-7.4) overnight at 4C.
3. PBS three times for 5 minutes
4. 14% EDTA for decal 3 days.
5. wash with PBS

For example, you have mouse sections, and your primary antibody is rabbit
anti-mouse
for your sample: dilute your primary antibody in your incubation solution (
normally PBST + 5%-10% horse/goat serum), followed by washing. then your 2
nd antibody will be horse/goat anti-rabbit antibody in incubation solution.

本人千老一名。最近实在是被折腾的走投无路了，想借助大家的思路，看看我这个情况
怎么办。最近几个月的实验总是出问题。我培养的细胞前一段时间总是被污染，好了一
段时间后最近开始大批的死亡，而且死得很怪异。比如一盘24孔板的细胞，有的死有的
长得很好。要是说是技术问题的话，一样的细胞一样的培养液怎么会有的死掉有的好好
的，还有就是基本上比较耗时间的实验的细胞全死光了，而且就是在关键的时间点上。
最主要的是细胞大批连续发生2次了，我和我们实验室的technichian们还有几个要好的
薄厚们讨论了一下都觉得不可能是我的实验技术有问题。我们一致怀疑是有人搞破坏，
因为不光是细胞大批死亡，还有就是westernblot也被人搞破坏，具体就不说明怎么被
破坏的了。我们怀疑的那个人是去年刚招的薄厚，是个中国人。他刚来实验室的时候我

清理不掉。Hood 最后是花了一笔钱请人来放了个熏烟弹，现在貌似好了些。
Incubator 我把所有能拆的东西都拆了拿去autoclave，还仔细的用bleach then
ethanol 把内壁搽了个遍，然后还是继续有感染！我都要烦死了。。。
一共两个多月，所有的细胞实验都被叫停了，耽误好多事情! 大家有什么建议？

之前也是总有真菌污染，而且实验室人太多了，很难防止某些不太注意的人。总擦也不
是办法，于是就买了这个incubator，之后就再也没有事儿了。
我们本来的incubator也是这个型号，只不过没有装UV kit而已，买新的同时也买了个
UV kit装到旧的incubator里头，现在都挺好。 |

然后用FPG这种能识别8-oxoG的酶来切，被切的基因组位点会产生nick，阻碍PCR
polymerase，基于这样的原理做qPCR定量来研究某段序列对于oxidative damage的反
应。
好像这种做法比较少见，大部分DNA damage的研究都是global的。但也有很好的先例比

发帖数: 183 | 15 最近在试RNAscope，ACDbio,用的是RNAscope® 2.5 HD Detection Reagent-RED
老鼠的皮肤石蜡切片。用了Kit里的Positive Probe, Negative Probe, 我的target
probe, 还有以前同事的一个已知work的probe也做阳性对照。试了4次了，
用同样的片子试几种probe，现在的情况是：
positive probe 看到强的signal，dots,就算work了。
target probe 也看到些红色signal,算work。
但negative probe 也出现一些紫红，虽然明显比target probe 弱，理论上Negative只
应该有Hematoxylin的蓝色。
用同事的probe的片子，很强的信号在皮肤的真皮位置（是期待的结果），但问题是我
的片子在皮肤的表皮，也看到一些信号，这个不是期待的，同事以前用这个probe做老
鼠的片子，表皮只有Hematoxylin的蓝色。
感觉Negative和同事probe的结果，出现的不想要的信号，可能是同样的原因引起的，

一个做投资移民刚到美国的客户A在国内有白蛋白的经销许可证(据说这个许可证很难拿
，不过他有)， 目前是从欧洲进口白蛋白到中国。他想联系在美国可以供应白蛋白的厂
商。他给了我一些资料，就是现在进口白蛋白到中国的美国厂家名字。于是，我作为他
国内医药公司驻美国的市场销售代表开始联系。
可是效果不佳。我联系了资料上的所有在美国的企业。凡是有email的我都全部去了
email，说明了合作意向。有两家公司我还打了电话过去，不过他们不接受电话咨询。
到目前为止，只有两家美国公司(CSL Biotherapies和Baxter International)回复了我

2013.7 发现第一版网站设计太复杂了基本不可能完成。转型，开始开发第二版，同时
线下开始卖钢琴
2014.2 第二版上线，总计卖出数百台钢琴，搅乱了市场，收到琴行的律师信
2014.3 修复了和钢琴的关系，卖出第一辆汽车，网站开始盈利。
心得体会：
别浪费时间申请incubator和参加比赛，大部分不值，尤其别申请国内的创业园。
最好一开始创业就彻底理解什么叫MVP。
好好做产品，别浪费时间找VC
好好做生意，别浪费时间找media
angel真心是angel
大概汇报一下。现在项目开始seriously融资，版上有做super angel的或者有认识的朋

because someone gotta make a decision for something you can't agree upon.

angel和VC），但一般较少(<2万)，要5－10%,不过看来能满足你的要求。
还有，别忘了找所在州的startup grant，一般也有1－2万。

这两天新闻报道里提到IBM或出售其半导体业务，不知道大家有谁注意到了这些报道里
提到的潜在买家中，有一家叫global foundries最有可能中标。大家如果开车路过237
和great america parkway，会注意到那里在建一片新的玻璃幕墙的大楼，差不多快建
好了。其中有一座上面巨大的logo，就是global foundries。他们原来的办公楼在237

- Rabies can only be transmitted by a rabid animal. A rabid dog or cat cannot
live for more than 10 days after it is infectious. So, if your dog is still
alive and well, since it is already 7 days later, most likely, it is not
infectious when it bits you and you will be fine. Though the incubation time
for rabies can be very long (months), a dog within its incubation period for
this disease is NOT infectious.
