normally takes up to a few hours to finish an ITC experiments. Not a whole
lot of enzymes are that lousy.
To do it right, you have to make an inactive mutant that doesn't perturb
substrate binding.
In M-M curve analysis, you can back up a term, Km, which is close to an
apparent KD. |

目前筛药assay大部分这几类
kinase/enzymatic activity
cell toxicity/death
cell proliferation
protein-molecule interaction
imaging-based high-content screening
这些assay可以是完全in vitro，也可以是cell-based，然而它们的readout都是一些细
胞或者生化指标，和疾病这个层次还有相当距离。 |

She published that Nature paper as a rotation student in Shi lab. The TET1
enzymatic activity Science paper was done as a grad student of Rao's. Then
she landed on NYU faculty position with doing postdoc!
Rao invested amounts of efforts in that Science paper - it is hard to
imagine it is just merely stealing.
This field is just like this - many thinking of the same idea and question.

在Lander的文章中，figure 2把CRISPR的发展划分成了三部分：

标 题: 哥来科普一下，一一公的剪接体还是有希望的

大分子照相相关的炸药奖加上这次的，已经发了14个了（见下表）。
之前主要是基于X衍射或者核磁共振的技术开发和应用，拍一些单分子的裸照。82，91
，02年颁给X衍射和核磁共振的技术开发，剩下11个的给了应用。
而冷冻电镜，这次算是第一个（可能是第二个），直接给了技术。用它可以看更加复杂
、分子量更大的大分子复合体。这些大个头的结构当然更有复杂也更有意思。
这个技术开启了一个新时代，细胞里重要的复合体实在太多。接下来可能拿奖的那就是
：真核细胞的细胞核通道复合体；一一公搞的剪接体（这个结构无论在生命进化和疾病
相关都非常的重要）；G蛋白偶联受体（跟N多疾病有关）等等。
彦宁的工作好像确实没有什么希望，葡萄糖转运蛋白跟上面这些不是一个量级的。

easy to try and not expensive.
