不是ssDNA,但怎么克隆一个selection marker进去，又不影响感兴趣基因的splicing,
我还没有想出办法。除非你的突变点靠近后面的UTR,而UTR又比较小，你才有可能加在
你的基因后面。
按我作high-through put的思维，你最好作dilution method,一个well里平均放100个
细胞，然后duplicate每个well, 一个提DNA,PCR，genotyping。再把阳性的well细胞（
duplicate)dilute到single细胞per well，在genotype。这样，你一百个well/PCR能覆
盖10000个单细胞，还没有，就该放弃了。 |

我们老板，要求我们每次qPCR，对每对primer都要用input的dilution做standard
curve。而且要用定量的input做standard curve （10, 1, 0.1, 0.01 ng/ul）然后
sample的定量就用这个standard curve。我觉得更合理的做法是，应该是根据实际IP的
样品量（1/20, 1/200, 1/2000 and 1/2000 of ChIP input）standard curve。这个问
题，跟他争论了好几次，后来就懒得跟他争了。
另外，至于就是是否需要每次都做standard curve？我觉得其实是没有必要的，甚至用
input做standard curve是完全不需要的。standard curve本身的目的就是为了计算
primer的efficiency，以方便定量。其实，模板的量，也会影响primer的efficiency，
primer对input样品的effficiency不一定跟IP样品的一样。而且，qPCR本身是有每次

For polymer in solutions, the calculation for concentrated and dilute sample
are different.
Typcially, for concentrated samples, the so-called structure factor will
pollute your scattering profile. (Well, some people may like this
structure factor if they want to study the interactions.)
For dilute sample, things will be much easier. However, you still need
to have an idea of the conformation of your polymer before the calculation.

pack得太紧，那哪里还有微米尺寸颗粒一说？呵呵。
如果说微米颗粒意味着可以忽略颗粒间的作用的话，感觉颗粒状的情形
应该会有更小的loss。在极端情况下，当颗粒的尺寸小于domain尺寸的
时候，这个就是一个superparamagnets的集合，忽略相互之间的作用，
基本上不会有任何hysteresis的行为（包括magnetization，其名也源于
此），也就不会有由此产生的motion和heat作用。
我觉得比较一下磁性-非磁性合金（显现铁磁性）与相同比例、磁性颗
粒镶嵌在非磁性背景的混合体的差别比较有意思。这几年dilute
magnetic semiconductor就一直有磁性杂质是否uniformly diluted
的争论：有人认为材料中磁性分子很均匀地、分子尺度地分散；而有些
人发现磁性分子形成cluster分布在材料中。我自己做的不是这个方向，
