nano material bind到病原体上，就可以测到特殊的spectrum |

等）。这个细胞团即使经过PBS多次清洗也是很松散的，感觉不是那种纯细胞团PALLET
样。我用这个细胞团进行BEADS ISOLATION,这一步基本要花4个小时以上，杂质经常堵
塞BEADS管。混合细胞里面杂质太多，多糖啥的，请问有啥好的方法清洗这个细胞（去
除多糖杂质）？
之前发表了几篇文章后，几乎每周都有一些不知名的杂质来EMAIL约稿。我自己想审稿
，给绿卡攒，这些杂质靠谱吗？ |

发帖数: 856 | 4 我这个问题很奇特，我用Invitrogen的dynabeads做ChIP-seq，最后拉下来的DNA量总是
比较高，做qPCR也没有enrichment，但是用同样的protocol，换回agarose beads，就
很少碰到这种问题。这不合理啊，dynabeads按理说背景要比agarose beads低很多才对
啊？！不知道大家有没有碰到这种情况？我试过好几种洗的方法和试剂，比如最常用的
就是低盐洗一次，高盐洗一次,LiCl洗一次，TE洗两次。也试过用RIPA洗过，也试过大
量延长洗的时间，但是最后elute下来DNA的量还是比较高。
另外，能不能请推荐一个比较好的做转录因子ChIP-seq的盒子或者详细的homemade
protocol？我做的这个转录因子，比较low abundant，可是我的材料是primary tissue
，又没法一次拿到很多的细胞（一次运气好的话可以有50-100million）。看有的文章
说现在10million就可以做转录因子的ChIP-seq了，可是我从来没成功过。现在真是快
被逼死啦。。。。快救救我 ：（ |

的人说服了就可以的。
前几年离开原来实验室做公司去了，留下一篇基本写好的manuscript，老板让接手的烙
印博后小兄弟做个co first author补点数据准备投个大文章。没想到接手的小兄弟不
好好补数据却去检验我做好的部分，过了些日子老板抱怨我的数据不可重复，因为跟老
板本来关系特别好像朋友一样，如果造假就像坑朋友的饭碗当然是特别严重的。所以特
别安排了一个礼拜的时间回到原来的城市，看看小兄弟究竟有什么实验操作问题，全程
烙印动手我就看看加动嘴皮子。
其中一个实验是endogenous co-IP，烙印说是对照的抗体也有pull-down和阳性的差不
多，而我的数据里面对照是干净的。抗体beads结合完了以后需要清洗，我看烙印把离
心机设定到8000rpm3min，当场叫停，这么高的速度做什么，我都是1500rpm20sec，抗
体beads这么大太容易离心了。因为这是常识，感觉都算不上技巧，方法里面有一些没

发帖数: 89 | 7 如果预先知道会有人不会洗beads，肯定把这个写上了。我自己原来也没有人仔细教过
，根据常识来说细胞只要1500rpm可以离心下来，这个beads放在那里几秒钟自己都能沉
降了可以看得很清楚，估计200rpm就足够了，我用1500rpm应该已经算是overkill，这
个范围可以很大应该不算关键参数。只是哪里能想到别人用8000这么搞。实验记录本已
经尽量详细了，要是不详细的话光改这么一个条件应该还是做不出来。每天工作量很大
，写记录只能把握一个尽可能的度。前面的Dua没有检查到自己头上不要轻易下结论，
很可能你自己的记录本也缺了点啥东西谁知道呢。
写这个没有支持韩春雨的意思，虽然希望韩是真的，假如韩的结果证实不行了希望不要
影响小实验室模式。 |

伤BEADS，这个也学对于新生不算尝试，但是但凡有心的实验者应该都不是问题。所以
实验有时也还真有点艺术，对于有的人复杂的东西也比较简单，但对于有的人，很简单
的人也很复杂...习惯细节决定成败... |

1）当年olins夫妇如何在in vivo 证明这个bead-on-a-string structure的？我的印象
是没有谁用fluorescent microscopy做了(resolution也打不到啊）。最近的in vivo
super-resolution也只是证明了nucleosomal clutch（发在cell上，大家自己找），没
有证明bead-on-a-string啊？哦，对了，biochemistry method, MNase digestion, 再
提取DNA， gel。OK。但是这个MNase digestion的chromatin不也是提取的吗？
避而不谈这个问题了。我实在想不出有什么其他biochemical method 去印证我观测到
的结构了。
2） 如果你来做这个研究，用什么东西来提取chromatin比较好呢？前提，无损higher
order chromatin structure. |

。准备pulldown histone, reversecrolink, 收集DNA做Dot blot.
直到SONICATION这一步都行,Chromatin reverse crosslink后 DNA在agar gel H3 在
page gel上都有信号。 但是pulldown后没有信号。主要基于 Farnham protocol。
他的ChIP buffer就是RIPA,beads也没有block,这个没问题么？有些别的protocol用
salmon DNA block beads,salmon DNA的methylation会影响 DNA methylation dot
blot结果吧？ |

10月份的时候，我开始试婚纱。先看了几本杂志，找到自己喜欢的型。再
从yelp上找review好的bridal shop, 打电话预约试纱。
第一个去的是david's bridal。明明预约了，跑到那里说找不到我的名字
，只能临时拉一个人帮我试。这个sales很pushy, 也不听我的意见。我说不喜欢bead,
她给我找的第一件全身都是bead。后面试的几件也没有完全符合我的意思。第一次试纱
很失败，对DB印象也很差。
第二个去的是个local小店，外表破破的，但是网上review很好。店主亲自
帮我试纱，我给她看我带去的杂志照片，她从店里拿出几件，件件都符合我的要求。我
有些不知道怎么取舍，她就帮我分析每一件的优点缺点。这里的服务比DB不知道好多少
，价格也不贵。
第三个也是local店，服务很好，纱纱也很pp, 无奈价格超出了我的budget。
在第二家店，我其实已经找到了the one，那种感觉很神奇，一穿上就觉得

，也算是通杀饵之一。
fly fishing的精髓就是要自己制作合适的flies，只有自己针对实际情况制作、调整
flies的细节，例如颜色、大小、重量等，飞钓才能真正管用、才能真正多搞鱼！所谓
“细节决定成败”。建议弟兄们尽量自己学着制作，熟能生巧。
完整的原版Bead head无力八哥（加重型），为做到精美和耐用，制作相对复杂，材料
也昂贵一些，具体过程和材料以前有帖子。有兄弟反映说一没材料或者材料贵，二是做
不出来或者做出来很难看。琢磨、试验了一阵子，搞了这么个简化版无力八哥：利用较
少的、廉价的材料，同时大大简化了制作步骤和难度，实际应用中经过测试，和原始八
哥的效果不相上下。
（以下流程和图片对应）
1：材料：#10或#12直炳钩，1/8寸金色bead head，橄榄色或黑色marabou（毛毛，如果
搞不到，也可以使用普通羽毛根部的软一些的毛），Palmer Chen（人造亮丝线，小号

前几天带两个兄弟（姑且叫他们老L和老C吧）去试枪， 两位都是网上认识，从未谋面
。 见过C的夫人， 热心爽朗，绝对是个对脾气的， 不是一家人不进一家门， 老C也错
不了。两位除了几十年前大学军训外也没打过枪。 前一阵子在Dicks一起买了Benelli
Nova Tacital， 还没试过。
Benelli Nova Tacital是18寸半的枪管， 不到我常去的室外碟场23寸最短管长的规定
， 不能去。 唯一的选择就是室内靶场了。 室内只让打buckshot或slug, 弹药成本高
了些， 好在他两位初次打枪也打不多。 靶场门口见面后发现两位都和我差不多年纪，
举止沉着稳重， 让我放心。 他们的枪袋、耳罩都是一样， 连擦枪工具也是一样，
刚刚从店里一起买的， 我笑他们一看就是一个帮派的。
