1. ChIP buffer contains SDS and SDS kills M2 beads (refer to M2 beads
handbook)
2. you never know how much antibody you used. 10 ul of M2 beads perhaps
contain hundreds ug of ab. and saturate your chromatin. you need titration,
but impossible.
you may want to try M2 antibody which is a monoclonal antibody and the same
as the one conjugated with M2 beads.

抗原抗体。 在抗原都是阴性的情况下，如果只有一种抗体，就是surface antibody是
阳性的，是受疫苗影响产生的； 如果是有多种抗体，包括surface antibody, core
antibody, e antibody 三项抗体都是阳性的，则是由于之前受到感染后自愈造成的。

2. crosslink your antibody to gel particles (CNBr activated Sepharose),
use the gel to pull down your protein,then use high salt(eg., 8M urea) to

target will have a higher molecular weight. However, the result can turn out

Monoclonal antibody is against one specific epitype of antigen. If
it works, it works. If not (for example, under immunohistochemistry
conditions, methanol, paraglutahyde...), it won't. Polyclonal antibody
is against a great variety epitype of the antigens. Some clones of
antibody might only work for western, the others might work for immunohist,

western检测不出来，试试regular fluorescent microscopy看一下，确认一下

work,work了做flow比confocal容易

reaction (HRP conjugated 2nd antibody). It should be fine to use at a very
low concentration.
After all, it is all up to the antibody. For b-actin, sure freeze and thaw

So for me, unless it is extremely hard to get the antibody, I woudn't reuse

最近试抗体，发现Pierce的这个活动很好，只要你第一个抗体符合它上边说的有新的它
与原本没有试过的应用（WB, IF, IHC, IP等等），你提交相关图片就可以免费获得第
二个同等或较少价值抗体；如果第二个抗体也有新的应用，提交图片，免费拿第三个；
以此类推，如果你运气好的话，可以循环下去。我现在正在提交第二个抗体的图片，马
上拿到第三个抗体。但是这个活动马上就结束了，12/31。如果感兴趣的可以试一下。
这个活动对于我这种做非主流模式生物，专门的抗体比较少的人很有用。只要尽量选高
度同源的多克隆抗体，做IF总会有些pattern。

which domain it is designed against, if that protein is really ablated. for
instance,if that protein has 20 exons, and you expect that the protein
after exon 10 is knocked out, but your antibody available is against the
exon 1. then, if it is a clean knockout, this antibody will detect a shoter
band than expected (i.e. from exon 1 to exon 9, but not a full length

我也不是专家，专家都在讨论Cui J和内幕，我贡献一下自己的2 cents。
你的staining 肯定有问题。Lyve1只表达在lymphatic endothelium上，不在
macrophage上表达。你如果把图贴出来，可能大家还能帮助你。没有图，我只能猜测有
2个可能：
1）你的F4/80antibody不好。F4/80 antibody有2个clone，一个是CI:A3-1，一个是BM8
。8M8 fix的条件比较tricky，不容易在section 上stain，FACS很好用。如果你用的是
8M8，建议改用CI:A3-1，或者仔细摸一下tissue fixation的时间。Lyve1只有一个
clone （ALY7 ），很亮也很好用。
2）即使你用错了F4/80的antibody，也不应该2个antibody co－localize，LN section

LZ did not give much information of his experiment, so what you listed
in below is also pure speculation.
Here I attach a pic what I stained with LYVE-1 in LN,
which is also inflammatory.
The red signal is LYVE-1, blue is B cell region.
Fat should not be problem in isolating LN.
I agree with you that there might be problem of specific staining.
I assume that in the LN of his model, here might be expansion of
