after you get the blood, test them before do the experiment,
usually 2nd bleed or 3rd bleed is better than the 1st bleed.
but sometimes 1st bleed is okay, depending on experiments.And
you can reboot the rabbit.
IgM is produced at first and diminishes, IgG is produced second
but still earlier than other three types, and the total amount
IgG is definitely much more than IgM.
Also, if your antibody is not enough, you can try protein A column
to affinity purify more IgG. |

you need much less if you are targeting circulating cell by i.v
depends on the first question.
if i.v., I would say 60-80 ug would be a good dose to start with.
It also depends on the antibody though.

去就行了 也不需要考虑哪个antibody好用 呵呵
其实拿bone marrow cell出来转了基因然后种回去已经做了n个临床实验了 效果不好说
但是至少是feasible的 |

In the last figure, they compare the neutralization potency and breadth of
the new antibody VRC01 with that of b12, a well-known broadly neutralizing
Ab against CD4 binding site of gp120. Under IC50 <50 ug/ml, VRC01 can
neutralize 91% of 190 Env pseudoviruses, whereas b12 can only neutralize 41%.
One question about the paper is that they design a molecule, RSC3, that
keeps the antigenicity of CD4 binding site of gp120 whereas remove the
antigenicit |
