，鉴于有网友问，特此发个帖让以后要想买lenovo电脑的网友了解了解，也许可以帮你

首先lenovo是不主动说他们有price protection的，我也是发现降价了给客服说我要取
消订单，理由很简单，才买一周还没寄出就降价$160，那我还不如取消重买一个（当时

找你要研究的基因的BAC，然后再E.Coli里面通过homologous recombination在
有兴趣的片段（当然要比较长的）两头加rare的RE site或者ZFN一类的，随便折
腾；里面再加点strong的aptamer, 或者变态点，加一堆lacO.
这玩意儿做成转基因IPS或者ES，好处是可以多copy, 十个八个没问题吧，更多
应该也行吧? 然后做成转基因小鼠，取你有兴趣的特定组织，或者细胞培养，
做ChIP的前面部分就是fix，洗掉细胞质，然后instead of sonication, 用RE
一顿乱切，配合aptamer purification. 你可以先size select然后affinity,
或者先affinity然后size select, 或者几种不同的方法series affinity，
anyway, 拿到东西decrosslinking上ms spec, 当然前面crosslink的时候不能
用glycine quench.

recombinant or semi-pure proteins) experiments are dictated by the
sensitivity of your detection method and the binding affinity. There are no
gold standard. Generally, you have to try around.
I am suggesting some reasonable start here. For high affinity binding events
(tens of nM or tighter), a few micrograms of protein are sufficient. If you
load everything into one or two lanes, coomassie blue staining should pick

一些粗浅的理解。
首先，药物分子水平的机理太复杂，结构能看的就是，药物小分子和一种蛋白在某个位
点上的相互作用有哪些。就这些信息，仍然受很多因素的影响，很多时候都不是致命一
击，比如小分子affinity低，占有率低，或结构分辨率低，就会有很多问题，如果小分
子太小，fit电子密度可能性有很多种，或小分子太大，不是所有的部分都有清晰的密
度，这个时候就很难讲，在这上面出错的结构有很多。结构不能揭示的药物分子机理就
太多了，药物怎么代谢的（往往很多药物代谢底物才是真正结合靶点的小分子），怎么
进细胞或怎么上膜，即便是跟靶蛋白，小分子进入和离开靶蛋白的kinetics，on rate
和off rate，进入binding site的整个process（first binding site，secondary
binding site etc）都是静态结构不能看到的，而这些信息很多时候才是重要的，比如
对于很多药物，其进入binding site的通道或过程往往决定其特异性，而不是结构看到

GPCR的C terminus都是结合arrestin重要的位点，磷酸化与否是arrestin结合的关键因
素，虽然有些例外。receptor和arrestin结合不稳定并不是指affinity，而是整个过程
非常的dynamic，传统的观念，也是这篇文章的point，就是arrestin结合C terminus，
然后再构象变化结合到G protein结合位点上。但实际上，很多receptor结合arrestin
但是不internalize，或者结合arrestin但是没有arrestin signaling，这些都不是这
个简单的模型可以回答的，实际结合过程可能更为复杂，不是一个单一或均一affinity
的状态，甚至还有可能需要其它蛋白的帮助。如果真affinity很高，这篇文章里也不会

recommended dosage for recurrent plugged ducts is 3600-4800 mg lecithin per

港又方便，很多宝宝用品都在香港购买。以为自称有购物天堂的香港，购物品质和服务
有保证，谁知道第一次遇到售后保修就让我大跌眼镜！
香港荷花母婴中心（Eugene baby）是香港比较大的母婴综合店，在香港有8家分店。去
年8月份，我们在旺角东新世纪广场荷花母婴中心买了一部Britax affinity婴儿车，购
买价格5175港币。购买时，婴儿车除了整车和配件外，只有一本厚厚的使用说明书，没
有保修卡。因为觉的婴儿车是消耗品，而且这么大的国际品牌，一般不会有质量问题，
当时又恰逢荷花bb展人很多，匆忙之下没有留意到这个问题。
我宝宝是2013年10月底出生的，现在7个多月18斤。除去坐月子不出门的时间，目前婴
