八卦？我当时就觉得paper质量不行。
另，听说做NuRD complex的牛实验室还写了negtive的comment，只是不知道为什么现在
还没发出来？ |

Tamoxifen, a selective oestrogen receptor modulator, has been used in the
treatment of all stages of hormone-responsive breast cancer. However,
tamoxifen shows partial oestrogenic activity in the uterus and its use has
been associated with an increased incidence of endometrial cancer. The
molecular explanation for these observations is not known. Here we show that
tamoxifen and oestrogen have distinct but overlapping target gene profiles.
Among the overlapping target genes, we identify a paired-box gene, PAX2,
that is crucially involved in cell proliferation and carcinogenesis in the
endometrium. Our experiments show that PAX2 is activated by oestrogen and
tamoxifen in endometrial carcinomas but not in

发帖数: 311 | 8 当年我也听说这一帮子做epigenetics的人要到cell那搞，等啊等啊，我都快忘记这码
事了。今天很诧异看到好几个人提这事，俺还以为只是小范围传播哪。看起来这次是就这么过去了，
不会公开闹出来了。看老尚以后的paper好不好发就知道影响了。

发帖数: 56 | 9 他的那篇cell确实有些让人意外。毕竟做NuRD的实验室很多，不大可能发现不了LSD1的
，除非是NuRD和LSD1的interaction很弱。不过强的相互作用可能早被人做光了。。。
剩下的就是模棱两可的，大家抓住hot的蛋白做做就是了。
至于那篇Nature，好像后来还是有不少文章支持，至少我觉得另外一篇Nature好像比较
支持他们的观点

发帖数: 56 | 10 我也没看出來，你说的哪里反了。或者有没有别人发表的文章表示质疑，至少我没有读
到。不过估计有人质疑也就那样了，我现在根本不敢相信任何别人的paper，只能相信
自己拿到的结果。

The Shang Nature paper kind of propose 1, PAX2 is a transcription target of
ER, 2. PAX2 is positively correlated with tumor growth.
However, the 2008 Nature paper says that PAX2 is a co-repressor of HER2 gene
transcription, which is tyrosine receptor kinase implicated in malignancy
transformation. I am not sure about HER2 amplification or any functional
roles in endometrical cancer. But still the two papers' conclusions seems
very different. |

I didn't read them again, but in my memory, these two papers tried to
address the same question: why chemotherapy drug,tamoxifen, suppressed
breast cancer cell growth, but promotes endometrial tumorigenesis. Shang's
paper showed that tamoxifen could trans-activate PAX2 gene expression in
endometrial cancer cells, and the activation of PAX2 resulted in the
endometrial malignance.. The 2008 Nature paper showed that PAX2 is required
for tamoxifen-mediated ERRB2 gene repression in breast cancer cells. It
seems to me that these two papers are consistent and tackle one problem in
two ways.

发帖数: 1125 | 15 JHU的ap不是真的，原来有一个他们实验室出来的人在网上说过，老尚拿到了jhu的面试
，但是没有拿到offer，貌似有其他学校的offer，但是他决定回国了。原因很有可能是
英文口语问题。
但是我不知道为什么老尚把那个简短的6个月ap经历放在网上，真是败笔。不过貌似最
新的cv已经没有这段了。
实际老尚还是很牛的，做得应该不比同期的jhu的ap差,他无需加上这段不存在的jhu的
ap经历来加强自己的背景。。。 |
