设计oligo的时候可以把粘性末端设计好，链接前可以不用做酶切。
我试过的一个protocol，非常简单好用：
1. oligos用TE or H2O溶解，浓度100 uM；
2. 磷酸化准备如下，37C一个小时, 70C for 10 min.
15 ul H2O + 2 ul oligo (100 uM) + 2 ul T4 ligase buffer + 1 ul TNK
3. 退火反应如下，95C 5 min in PCR cycler，RT for 30 min.
18 ul H2O + 1 ul oligo F + 1 ul oligo R (final 0.5 uM fragment)
这个产物可以直接用于连接。

What kind of cells, which fatty acids are you using? Please give more
details.
Here is what I used for hepatocyte.
Preparation of the fatty acid stocks
The free fatty acids are prepared by dissolving each individual free fatty
acid in 100% ethanol. Most are readily dissolved in ethanol, though some
require heating to achieve fully solubility.
A final concentration of 100 mM for each of the fatty acids as follows:

1. Seed 1.5x106 293T cells per 10 cm plate in 10 ml growth media (
DMEM + 10% FBS). Usually, one T-175 flask of 293T cells at 90-95% confluency
is split to 12 plates (10-cm).

我是引物相同的都先混在同一个1.5 ml管子里，其实也就是俩管子，一个是
housekeeping gene another one is the target gene，然后按模板在分出来，一般
technical control，就是同一个模板3个重复，96 well也就是32个小管，16 each for
the two genes，然后模板加3 ul，但是一般是做4份的量，因为分装第三个总会不够
，所以要多作出来一份。我是10 ul体系，所以就是做40 ul （36 ul master mix+4 ul
template），还有这样你模板不会只加1 ul，相对而言加的量更精确些。不知道说明

作为Inventor，你一个人是无法迅速marketing新产品的，你需要找到和合作伙伴。
Small Home Appliance基本被几个大的品牌垄断，大的Box store你就不用浪费时间了。
As seen on TV是推广新产品的好办法，但是初始投入很大。你目前连投入大概不到1万
USD的电器的UL都不愿意做，拍短片比UL认证贵多了。
另外，没有UL你就投入生产好像不对，UL认为产品需要修改怎么办？
