我以前读一些paper,说RNAi knockdown 对老鼠的问题在于transgenic mice对于
heterozygou 非常耐受，就是说knockdown 50%都不会有明显phenotype. 而RNAi 在体
外也就70% 的knockdown, 所以构建transgenic line没啥phenotype,不知道是不是这个
原因以前一直没有line 出来。
我做transgenic RNAi老鼠主要目的是研究成体的KO phenotype.当然，reversible也是很重要的原因。 唯一办法就是构建XXX
-rtTA；teto-shRNA 来在成体knock down. |

Without knowing the upstream or downstream sequence of your integration site
, how do you design the primer outside of your transgene?

you will be able to design anther primer using known sequence. However in
the case of a transgene, it is no as simple as 5'RACE since very often the
transgene construct forms concatemers which greatly increase the complexity
of real products. there are several of these similar techniques such as "Pan
PCR", "selective circularization" or sth etc.
There are also commercial kits available.

signicantly increase the expression level of genes. the classic example is
beta-globin. w/o intro its cDNA barely expresses in cells. with an intron
even an irrelevant intron (insulin intron for example) it expresses at very
high level. you may wanna search with key word "intron, expression (beta-globin)
but i don't know much what's the situation in transgenic mice though. |

when you design primer to sequence sth, you don't need a pair of primers. So
if I design a primer from the head of insertion and go upstream to sequence
the unknown, I only need to know 100 bp sequence. then based on this 100 bp
I can design primer, one primer on transgene and one on upstream, whose
product is cross the insertion starting point, so to distinguish homo vs
hetero.
But I don't know whether the sequencing pcr can be done on genomic DNA.

RACE...I don't know but I want to believe there is a way to do this since
the insertional mutagenesis has been

STOCK Tg(CAG-cre/Esr1*)5Amc/J. 我set up 了5个mating cage ,都是Cre hetero X
WT mice, 交了3个月。子代genotyping后，发现还有Cre transgene 的数目很少，一般
12个老鼠，只有2个带Cre. 比例才1:6,不是理论上后代应该有1：4含Cre 么？
请教各位，这个比例是正常的么，怎么提高子代Cre的数量阿？

下面的论文发表的时间，是中国两种转基因水稻一种转基因玉米“终于”获得安全证书
、并引起全国转基因大辩论的时候：2010年2月。
让我们看一下摘要的简单译文（全文未见）：
“用近红外反射法，气相色谱-质谱法、高效液相色谱法、电感耦合等离子体原子发射
光谱法，并以化学计量法研究三种（分别）可抗霉菌病和抗虫的转基因水稻中重要的营
养物质成分组成（按欧共体标准），发现其营养成分组成和物理性状有非预期改变，其
中蛋白质、三种氨基酸、两种脂肪酸、两种维生素及其他数种（微量）元素出现了非预
期的构成变化。差异幅度：氨基酸20-74%，脂肪酸19-38%，维生素25-57%，（微量）元
素20-50%，蛋白质35%；植酸没有显著变化。相比于非转基因水稻，转基因水稻的营养
成分构成和物理性状的非预期改变，可能与基因的转入有关，阐明变化后的作用需要经
过更多研究。”
需要说明的是：这项研究所用的上述几种方法，全都属于常规分析方法，不是专门检测
转基因成分的高新技术，因此这项研究似乎不是针对转基因成分的（网上尚未见全文）

这些研究结果都没发现转基因比传统育种的更risky。
