真不觉得建tissue bank是建急需的事。只有当国内要用tissue bank的人到了一个相当
的数量，建
tissue bank的呼声自然会高涨，自然会有行动。国内的medical research也好，
pharmaceutical industry也好，现在有几个人用tissue sample？在需求不大的情况下做
database恐怕变成形象工程的可能居多。
清华多找了几个做结构的不是什么大问题，施一公主导当然会偏结构一些，就像王晓东
会偏生化，饶毅
会偏神经一样。而且，清华新招的人也有好几个做结构之外的方向的，随着招聘人数的

发帖数: 15 | 8 哎，都博士二年级了，还问这样的问题，真不好意思
我是用qiagen blood & tissue kit 提brain tissue的DNA，~ 20 mg， handbook上面
说 brain tissue的话25mg 的DNA 产量 15 ~ 30 ug， 那理论上我应该得到12 ~ 24 ug。
但是我做了几次，最好的那次是200ul elution buffer，浓度 ~ 30 ug/ml, DNA 总量
是6 ug。
今天又做了2个sample，200 ul elution buffer，浓度只有10 ug/ml, 再用新的100ul
elution buffer, 浓度~10 ug/ml， DNA 总量大概只有3 ug。跑了一块胶，可以确定是
有DNA的。
实在想不出来是什么原因。 我disrupt tissue的时候是用23 G的needle做的，然后加
proteinase K消化了6个小时，液体很澄清，应该不会是消化的不好吧。很简单的实验
，就是做不好，哎。。。。
Sample比较珍贵，不敢再做了。。。希望大家能帮我一下啊，谢谢大家！！ |

1. CreERT2 (就是第二代的CreER). Cre driven by tissue specific promoter.
我研究骨头。打了tamoxifen （我用oral gavage)10天后就有phenotype。但是
tamoxifen有些副作用，比如增加bone mass.所以需要optimize dosage.另外我用打了
tamoxifende wt mice 作为control,这样可以排除tamoxifen effect.
2. 我用过Tet-Off system. 需要三只老鼠：tissue specific-tTA; TetO-cre,加上你
的conditional 老鼠。喂老鼠doxycyline-containing water or food 直到你想要表达
基因的时候。
3. Teto-ON system:tissue specific-rtTA; TetO-cre,加上你的conditional 老鼠。

100%取下来，最后的结果control/treated才可以比较吗？
：：理论上应该取100％，但是因为常常细胞数太多，可以homogenize之后跑同样的
percentage。比如都跑总sample的10％
flow cytometry最后的那个结果,反映的是某种polulation的cell的分布百分比,这个概
念对吗？我们每次杀动物的时候，那个tissue都很100%难取下来，每只动物都只能取下
一部分。
：：反应的是某种population 的百分比。尽量多取吧，如果tissue是均质的，你也可
以只取一部分，比如如果是肝脏细胞，但是如果你想看的population不均匀分布在
tissue里，就最好都取，才能最客观的反应在总的population里的百分比。当然你可以
不必要都跑完，原理如上。 |

--这个不一定，只要是均质的组织细胞，取够染色的细胞量就够了。因为如下所述，最
终得出的是各个细胞所占的百分比。
****需要注意每个sample染色的时候ideally是1个million，细胞太多抗体量不够，BD
抗体一般是按着1微升/1million细胞/100微升体积里.
flow cytometry最后的那个结果,反映的是某种polulation的cell的分布百分比,这个概
念对吗？我们每次杀动物的时候，那个tissue都很100%难取下来，每只动物都只能取下
一部分。

ZL1999 (胖头猫)提到了confocal和一般fluoresent microscopy的区别，除了有一点我
不太认同，根据我的理解，confocal microscopy的photobleaching问题应该比一般的
fluoresent microscopy要小，因为前者illuminate smaller volume of your sample。
如果你要做的是培养的细胞，你可以不用改protocal，我一般用8-well chamber slide

tissue,我认为其dynamic应该是不同的。fast turnover tissue 来说，quiescent
stem cell-->proliferating TA cells——》committed progenitors_-->terminal
differentiated cells 这样的过程有利于stem cell快速扩增。但对于很多slow
turnover tissue, 比如，肌肉，骨，这样的TA population在生理状况下是检测不到的
。当然对slow turnover tissue的 stem cell dynamic的研究本来就很少。
关于multipotential是否该是stem cell的定义，最好的回答是Pax7+ satellite cells

大家都管自己的细胞叫干细胞，实际上只不过是炒作概念，
做development的都说自己是做的干细胞了。

vitro筛选的效力惊人，只要有足够的化合物，in vitro活性都可以得到。
真正的瓶颈在in vivo筛选，这里面的最大问题的动物模型，现在最常用的是大鼠和小
鼠。这些动物in vivo和人类的相关性、预测性等完全没有办法，动物模型本身的病理
状态和人疾病的相关性等。如果哪一天猴子模型可以很普遍的应用，可能这些预测性和
相关性会好很多。

我做In situ hybridization，设计了3对probe（500bp左右），3对proble都是发现
sense和anti－sense有相似的pattern。
我一开始觉得是non－specific binding。后来越做，我越觉得signal是对的。我做了
transfected和non transfected cell pallet，只有transfected有信号，而且sense和
anti－sense都有信号（sense弱一些）。
因为我的基因有的tissue表达，有的tissue不表达。我又多做了很多tissue，发现我的
probe有表达的tissue才亮，说明probe是好的，看起来信号也挺好。
可是怎么解释sense和anti－sense都有信号的问题？有ISH的大牛能帮忙trouble
shooting吗？ |

染人的细胞。对此新发现，武汉病毒所还把它以武汉病毒研究所的英文简称命名“WIV1

dead tissue not involving bone, tissue debridement might work. You could get
a2nd opinion from another general surgeon. Walk in a general surgery office
and tell the front desk it is an urgent case and need immediate
reevaluation. Bring all her lab/X-ray results, and make sure you understand
what they said.
Bottom line is that dead tissue must be removed since infection can spread
out.
