isn't mounted now. And anything you create may result on the / partition which
contains that. what's your /etc/vfstab, and which partition you have problem?
Strongly suggest you don't touch anything until find someone to fix it.
I heard that there are tools like undelete/hard disk recovery, but it will
create very huge number of data (200M original will let me few Giga row data)
important stuff need back up, not recover |

the CD, then can not see D drive. Only see C drive for 100 GB. Have checked
all the drivers are installed correctly. HD is a SATA drive.
Any idea how it is? Need the computer for paper submission, please help! |

我的protein sample是cell lysate, 浓度大概是40ug/ml,总共3ml左右.想concentrate
到2mg/ml左右。试了millipore 的Amicon column, 是可以concentrate到想要的浓度，
但是回收率只有60%左右。大概很多protein粘在了膜上。因为最后的蛋白量对我很关键
，请问有没有什么方法可以达到80-90%的回收率？

Amacon是好的浓缩纯蛋白的方法，对cell lysate不是太好。首先，有个分子量Cut的问
题，膜孔径的大小。其次，小蛋白本身就会丢失。自然你的recovery 不会高。
1.Salt cut，复溶后小心dialysis，去盐。蛋白还是活的。这么处理后，能否到80-90%
我也不知道。
2. methanol precipitation，蛋白应该是变性了的。
可能还有别的办法，期待大牛们讨论。
