purification变性条件来第二轮纯化（此顺序是实验需要，不要问为什么）。目前结果
是第一轮IP很好，但是His-purification后蛋白都在unbound fraction. 我用的是
Qiagen Ni-NTA Magnetic Agarose bead,据说适合纯化含量比较少的比如mammalian
cell的蛋白。IP后的蛋白都经过 buffer exchange，也没发现有与Ni resin
uncompatible的成分。请帮忙trouble shooting。会不会因为蛋白浓度太低导致与Ni
resin结合不好？His-purification有没有target protein concentration threshold?
是否需要改用binding capacity更大的Qiagen Ni-NTA agarose (不是magnetic bead)？ |

I am doing purification of membrane proteins to get enough concentration for
NMR study, the protein was labeled by his-tag
Firstly, the protein was expressed by E. coli, lysed by several enzymes,
agents and detergent(empigen) to solublize everything, then it was purified
by Ni-NTA column, the protein shows at the right MW(about 18KD) at commassie
blue stained gel.

继大前天录制了舞动奇迹的主题曲后昨天又在长沙拍摄了这一季的宣传片 一个下午的
拍摄蛮顺利的很多动作虽然没有音乐辅助但还是按照导演的要求在想象中完成了 相信
卫视一贯高水准的后期制作会把宣传片做的很好 当然也少不了两位男主角的卖力演出.

如果装了,还要装在厨房sink下面的那个净水器吗?
我的理解是软水,是去钙类的硬水的东西,但是净水要去杂质,可以饮用. 这搞的我有点
晕,不知道怎么research了.另外市面上最好的牌子或是主要牌子有哪些? |

发帖数: 59 | 23 忘说了，我就是用变性条件做His-purification的，用8M Urea, 100mM Phosphate,
10mM Tris, pH7.8做binding.
我确定His tag是表达的（based on size and DNA sequencing)。基于很多原因，不想
把 his-tag放到另一端。如果是变性条件，tag就肯定不会被包埋吧？
还有另一个可能，之前IP elution 有用1%SDS，之后做buffer exchange (Milliopore
Centricon)后可能SDS去除不完全。但是最坏可能也就是His-binding时有0.1%左右的
SDS，看Ni-NTA compatible list 应该不会影响binding啊 |

his-tag 应该是wesetern和purification都可以。
GST的话做纯化可以，做western，由于有点大，可能会对功能有不可以预测的影响。 |

I am in Boston now but I need to relocate to Maryland since my family is
here. I have been searching jobs in Maryland and D.C. for a while but found
it is really dissapointing that very few pharmaceuticals industry is here
and few position is open. Is there anyone here knew any research company
