Broad和CSH这两个source的distributor，主要就是pLKO和pGIPZ两种骨架的shRNA.两种
我都用过，因为每个基因一般会order3-5个hairpin，基本上里面至少有一个的
knockdown是不错的。 |

看到smear，然后切下比如450-500bp的区间做PCR。但可能是50bp的interval太小或者
自己技术不好，所以最后PCR出来的亮度比较低。（当然也不排除ligation efficiency

还没拿去测浓度。只想问问有经验的前辈这样的情况能拿去测30 coverage的WGS吗？
另外还有一个问题，就是，我们切胶的smear每一个区间（300-400，400-500，500-600
）这些区间里都一定cover了genome的每一个部分吗？我总担心genome的有的部分没有
被涵盖到，那岂不是就人为引入了很多bias？

然后做serial dilution,做标准曲线。
一定要用PCR产物，质粒不可以。用质粒结果极其诡异。
我自己做的library，一般10ng/ul, 总量100-150ng，Bioanalyzer还算准确。
如果能自己用bioanalyzer最好，如果是Core做的话，一定要注意峰的面积是怎么算的
，那个分析软件有时候很crazy。 |

要求只需要提供函数值，不需要计算gradient的，然后就是要求能够有box-bound
constrained optimization。我只会用c++，所以最好是c++的库，其他的库能够用c++
调用的也行，对了，必须是multidimensi的。
其实是要optimize一个微波电路的各种参数，希望模拟出来的电路测出来的东西和实验
结果是一致的。
