网上搜了下，都是要用土豆自己做。
马铃薯葡萄糖琼脂（PDA）培养基是分离菌物和细菌的常用培养基，其做法是称取200g
马铃薯，洗净去
皮切成小块，加水1000ml煮沸半个小时或高压蒸煮20分钟，纱布过滤，再加10-20g葡萄
糖和17-20g
琼脂，充分溶解后趁热纱布过滤，分装试管，每试管约5-10ml（视试管大小而定），15
磅蒸气
（121℃）灭菌20分钟左右后取出试管摆斜面，冷却后贮存备用。
请问这里有买现成的原料自己配吗？就是用 葡萄糖，agar 和（土豆粉？）按比例混合

seen in 5-7 day. If it still does not work, change the mediumn every 4 days
or order new cells from ATCC. We routinely do it and no problem found.
2.It has nothing to do with stem cells. Most stem cell medium are serum
free. But Soft agar uses 10-20% serum (I even saw people use 30% serum for
weak oncogene transformed 3T3.).

Ab resistant colonies on the Ab-selective plates.
I am wondering if only the lysogens can carry the mutant gene with Ab
resistance marker.

香蕉啊，酵母粉啊，氨基酸啊，加上agar什么的煮了调来调去而已，貌似没有
加酒精。所以在我的印象中，果蝇对于不含酒精的食物也是会愉快进食的。
含酒精的水果什么的，貌似是他们产卵的首选地，倒不一定是进食的原因。

应该不是因为省电的问题，实验室再省也不会不开shaker, 那点电钱相对于
实验室材料的钱和你的工资来说简直就什么都不是。
我猜他们应该是担心你不懂干活乱做实验，把agar plate （？） 放进去摇。
稍微解释一下就好了。等你一些初步结果出来之后，他们知道你是能干活的人，
一般来说应该就会好一些。

白斑的比例还可以，就是总体数量低。
3. 确定是42度。这个也只影响白斑数量吧？
4. antibiotics没问题
还有可能是什么问题？LB agar两年前的。但两个月前克隆另一个基因还很好啊。

。准备pulldown histone, reversecrolink, 收集DNA做Dot blot.
直到SONICATION这一步都行,Chromatin reverse crosslink后 DNA在agar gel H3 在
page gel上都有信号。 但是pulldown后没有信号。主要基于 Farnham protocol。
他的ChIP buffer就是RIPA,beads也没有block,这个没问题么？有些别的protocol用
salmon DNA block beads,salmon DNA的methylation会影响 DNA methylation dot
blot结果吧？ |
