there is usually a large RFE (Request for Evidence) asking for lots of proof
that the company exists and has work for the beneficiary. They want to see
the lease for the office space, a copy of the floor plan showing where
employee workstations are, photographs of the premises, contracts for work,
payroll records, federal income tax returns and quarterly returns, etc.

hire, fire and control the employee is a factor. So in a 2 person startup -
- this may be an issue depending on the equity split.
Second, the position must be considered a speciality occupation --
typically most startups are in tech and the positions they tend to hire for
are H1B caliber, but you still have to go through the analysis.
Third, the startup should reasonably need a full-time X (developer,
engineer, etc) at the stage they are in. USCIS may argue that you should
have just hired a contractor or a part-timer. Ultimately, this may depend on
the company's current plan for growth. A business plan with projections for
hiring, cash flow and growth is useful.
Fourth, the company must pay at least the prevailing wage. This is
determined by county, position and level. If part-time, this will be an
hourly rate.
On the note of part-time versus full-time, sometimes, it may make more
sense for a startup to hire a part-time H1B rather than go straight to full
time based on reasons discussed above.
Finally, money helps. While technically, for H1Bs the company's "
ability to pay" is not a requirement-- in practice and when applied to
startups, it does matter. Funded startups have better luck getting approved
and overcoming nasty RFEs. It helps to have well-known VCs or accelerators
on your side. Having Y Combinator or 500 Startups back you is a major plus,
as far as I'm concerned. The track records of the accelerators, VCs and
startup founders are often taken into account.
