信号通路的影响。我的问题是有哪些蛋白positively或者negatively调控endocytosis
？没做过endocytosis。我想用siRNA或者inhibitor（如果有的话）改变endocytosis
baseline，然后再研究我这个受体的行为。谢谢。或者推荐几篇参考文献也行。
还有我不确定这个受体蛋白进入细胞是通过传统的endocytosis，那如何分辨其他的机
制呢，比如pinocytosis或者caveolae？ |

的readout，合作者都找好了，但老板不同意这么搞，所以我现在只能逐个分子摸索着
往前做。不过我这个蛋白进入细胞的途径应该依赖于endocytosis machinery.

polymerization. You can differentiate these pathways by combinging knockdown
clathrin/caveolae and actin polymerization inhibitors. |

按你说的就是用生化的方法分离出caveolin-1 fraction和no caveolin-1 fraction (
clathrin)，然后western blot这些分子对吧。我分离过ER, nucleus, mitochondria,
fractionation还是挺tricky的。

basically, if you have pellets from 10cm plates, resuspend the pellets in
500ul 500 mM sodium carbonate (pH 11), sonicate (3X/15 s). The lysate is
mixed with an equal volume of MES-buffered saline containing 90% sucrose and
centrifuged on discontinuous sucrose gradient (45%, 35% and 5%) for 16 h at
175,000 g and fractions will be collected for analysis.
