板上各位PI, 我很想请各位办一个系列座谈, 可惜没有能力.
各位已经坐上教授位子的, 能不能说说怎么从postdoc成为PI. 如何做一个好的博后/学
生已经讨论的很多很多了, 大家心里其实都清楚. 可是除了发好文章, 那些有志于成为
PI的博后该如何努力积累资本, 该建立怎样的能力, 来成为PI, 很多人都不是很清楚.

各位已经坐上教授位子的, 能不能回顾一下, 你觉得你当年有哪些(能力/资本)比较出
色的, 哪些还不足的? 说出来对大家都会有帮助的.
我想我没有资格讨论这个话题, 因为我才开始postdoc没多久. 但我很想听听各位的意
见.

Pretty much my case...
All my peers quit. So when finally there were some openings in my field, I
was kind of the only one left...
In my boss' wise words:
Getting a job has to do with market fluctuations much more than
whatever you do.

Good pubs is important, as you mentioned. An equally important factor
is your research direction – what you do. In the faculty job market, the
hiring department cares more about what they need, which is often in
line with what can bring in funding from the NIH.
Current hot topics include stem cells, small RNAs, genomics, epigenetics,
neurocircuits, systems biology, ... etc. Of course, the high-in-demand
research areas may change once you are ready to be on the job market.

You shouldn't worry about connections now. You establish yourself
by publishing high-profile papers. Then others will be happy to

me too, have asked quite a few bulls, different people have very different
perspectives about this...I always
feel it is very important but most of answers are not for postdoc...it seems
more about pedigree/camps less
about the network...

The important component of "connections" is just name-recognition.
If others know you do good science (because you published in CNS),
they would like to invite you to give a talk, would like to collaborate with
you, and so on. That's how you get connected.

My boss is a young faculty. He entered my school in the same year as i
did.
He launched his job with the help from his Ph.D. advisor, who is a big
bull in my school. My boss is brilliant and super hardworking, but with
only PNAS and no CNS

so do all (most of ) folks from your lab land a job in a top place? Or has
ever anybody got kicked out? BTW,
how are things going?

Sure, but what about before one had CNS...and say the MS, grant reviewing
etc etc...in our case, many/some of
the reviewers are either my boss' friends or from a same lab...and also they
had tri-lab annual retreat with
peoplem's lab and another big lab in stanford, so when I look around, it
seems they would virtually know half
of the people in the field...also, how important/critical to write reviews
as postdoc or junior faculty? That
seems a big boost for some...

regarding top place, I would say 50%. 20-30% went to decent place. remaining
20-30% go for industry and other careers. If you don't do anything stupid,
you will not be kicked out. Things here are going slowly but fortunately I
got an internal fellowship,quite a relief.

I agree.
a hot area + a supportive big boss + numerous quality papers + good
presentation and personal skills = faculty position.

You have good publications, and you are in a hot area. If you can make your

俺引用某大佬的教导哈 年轻老板 接reviewer活勤快点 写review积极点 干公共活比如
study section啊之类的多一点 一不小心就成了super star了 哈哈

本版近来铺天盖地的对中国 PI 的恶毒谩骂, 已经让不少 PI 对这个版敬而远之了. 估
计参加这一讨论的 PI 的数量会很有限.

I think connections are not that important, but collaborators are. If I
have 5 good collaborators, that's more than enough.
I have a particular style of doing science. Might not work for everybody.
I typically just pick a problem I like and attack it with latest tools.
If the tools are lacking, I invent them.
I almost never pay attention to what's hot right now. In fact I avoid
them. As a result, there are really not many people for me to connect
with. I typically find that people like to connec

Most people who ask me for reviews are my fellow grad students during
PhD.
I don't like to write review papers and giving too many talks.
But again just my style. I like to hide for a while and connect with
nobody, then come out with a big paper once every few years.

well, u are in a wonderland, although I always like to have ur perspectives
but yours never counts, to me:))),
shall say applicable to me...Yeah, seriously I agree with you, that would be
what i would love to do, but I just
don't think i could for a number of reasons...science to me still somewhat a
way of making a living although I
hope there is such day it would be just my luxury hobby and I can do what I
am most passionate about...

well, if I have another ten ys to fool around, maybe I would/could do that:)
))...I think my boss would be alright
in the coming ys but a little cow or not would depends on so many factors...
I am the only postdoc so far...it is
so hard for us to get good postdoc, my boss is a bit picky but getting less
and less picky...some we want but
they don't come but the others we don't feel would be a good fit...it is so
hard for a junior lab although the
boss is very decent and the field is filled with p

做几年后的当红方向最重要。牛校牛 lab 都是加分用的。
当然,如果赌错就郁闷了。这就是为何地球上千老这么多的原因。
这年头如果不 ride the tide 要找个好 faculty 位置都不简单。不是说完全
不可能,但是进好学校好 program 的绝大多是追热门方向的。我也崇拜那种不管
别人做什么都千山我独行的牛人,但是这种人的存活率在现在实在不高。现在不是
1960 年了,你只要脱离群众一下 funding 就立刻成问题,junior 的 faculty
找不到,你连 career 都不会有。

I just think it's impossible to predict what's going to be hot in 5 years.
So I just follow my heart.
If you publish a good paper, then what you do becomes hot.
But I use the latest tools, I think this is important. I think what
becomes hot is largely what's doable, which is tied to tools.

希望你喜欢的方向几年以后会变得很热。不然梦想破灭的时候会非常,非常痛苦。
其实做非热门方向的人心里也都知道找 faculty 会不容易,但是大家会集体
处在自我催眠状态 -- 觉得那个不幸变成千老的一定不是我。我一定会拿到 top
大学的 offer, R01, 从此过着受人尊敬的日子。
dream on. 还是做个热门方向实在。NGS, network, ChIP, whole genome
screening/profiling, 那个热干哪个。那个 DrBiologist 提的超烂方向
(codon usage)我还真作过一阵 -- 想看看它和 protein interaction
有没有关系。结果就是悲剧。前面那个叫人 submit 500 个 applications
的纯粹就是浪费时间 -- 你烂撒到 500 个你老板保证和你翻脸 -- 你丢的起人
他可丢不起呢。

你的 sampling method 有一个问题:你只听了 positive cases. 做过 classifier
的人都知道,你的 training set 里头一定要有够多的 negative cases, classifier
才会准。你应该也同时找找一些冲击过 market, 但最后灰头土脸的千老。

上面各位都说到热门领域, 我有个想法: 我不能预测什么领域是热门的, 但我可以大
体相信我的大研究方向在今后20年都不能很好解决, 比如肺癌.
这样, 我就想用比较新的方法去做, 这样结果和数据看上去比较fancy.

我 2002 年进 protein structure prediction 这屁行当的时候就是这样想的。
这鸟问题大家已经日了 20+ 年了,结果唯一的结论是 homology modeling 有用。
这种 long-standing 的问题大家会审美疲劳的。不中。

很多学校招人的时候看这个,如果你能说你是哪种方法的专家,比较容易找工作。

That's something good to have. But if you don't, there's not much you can
do about it. Just like somebody has a powerful dad and of course he has
better "connections". If your dad is a common person, you just have to
rely on your own hard work until you become recognized.

When I just came to US I did folate-targeted drug delivery. We chemically linked toxin to folate with some chemistry that the linker is easier to break in cells. Some cancer cell overexpress folate receptor on cell surface so the folate-toxin will enter cell by endocytosis. However it is technically hard to get the folate-toxin break efficiently to kill cells.
I transferred from Purdue after 1.5 years for other reason. You can check Philip Low at chem dept at Purdue. Also check endocyte.com

Thx for the info. the major problem w/ targeted delivery seems to be the
targeting ligand. it's really hard to find a targeting ligand that:
1. bind to a surface marker that is a very specific disease marker
2. have good affinity (nM preferred)
3. can internalize efficiently (usually means that the marker it binds
internalize constitutively).
I really want to find a senior cancer researcher to get some insider info
about the authenticity and expression profile of all the cancer surface
biomarker

I guess you are talking about endosomal escape? or chemical conjugation
simply
abrogate the drug property?
I'm hooked with the simplicity of conjugating aptamer with siRNA. both can
achieve certain level of specificity; both can be synthesized in a single
run; the toxicity profile of ONs are widely available due to the gold
rush in anti-sense field... Yet it's a shame that you only have that few
aptamers to choose from (well, only 1 or 2, according to my knowledge

my phd study focused on lung cancer mouse model and miRNA. We used LNA (
collaborate with Santaris Pharma) to treat cancer cell and lung cancer on
mice. But I cannot talk about detail at this time. The ON treatment field
is promising but with some difficulty to overcome.
