发帖数: 3778 | 43 个人建议哈。这个Amaxa最好还是先做个test。找个GFP plasmid，用他们提供的那个也
可以，然后买个test kit，里面有两个还是三个不同的buffers，然后按照他们的
manual各种情况都做一下。然后flow，这样可以有个比较准确的了解trasfection
efficiency。flow的时候可以加上PI，这样可以了解viability。
这样可以选个比较好的protocol。
如果选好了，这个Amaxa的效率是非常高的。 |

7-AAD应该是好的,我的293T细胞从培养皿detach后立即用7-AAD染色,viability挺高的,
有95%左右吧。但我用抗体染色时间长，要3-4小时，最后作FACS，7-AAD+ 的细胞就达
到了50-60%。
在室温下温育的时候， 死细胞和活的细胞明显分成两个population， 死细胞FSC明显变小了。
我的PBS是从公司买的， 1X，但从来没有测过pH。

从人刚开始建立小鼠纯系的时候，无意识的就以reproduction和viability来选择小鼠
，所以能活下来的都是一些特定的基因组合（strain），就这样也有些strain在某些方
面讨某些领域的厌。
人开始研究lifespan的时候并没有进一步去把这些老鼠怎么样。
所以我觉得wild mice值得研究，但是不能就此夸张的说lab mice 的data没有意义什么的

1. what is the promoter? if you know the promoter, you can guess whether it
is a tissue specific gene.
2. what cell type in pancreas express this gene, epithelial or other?
immuno staining should answer this question.
3. select cell lines based on answer to question 1 and 2
4. take highly malignant cancer cell lines, overexpress this gene by
transient transfection, cell viability assay, tumorigenecity (in vitro and
