如果是用antigen－affinity purification就会好很多。只是有时候那个his－tag还是
会干扰。一个师兄教我先用一个带HIS6的无关蛋白把non-specific的先去掉（比如
sunnyday最喜欢的actin就被我加了his6来干这事），然后再来做antigen－affinity。
如果是intein－tag提出来的这一步都可以省掉，只是intein得到的蛋白量好像没有his
－tag提出来的多。
要提到的是这个protocol在我手上得到的抗体浓度很低，我一般用1：100稀释，但是很
特异。还有就是不太适用于IP。
这个方法不用挂柱子，跑SDS－PAGE，直接转到膜上，block之后先把血清和actin－

你的蛋白很可能是以oligomer存在的（当然，根据我们的课本知识，gel filtration不
光是size有影响，还有structure。。。）注意到你buffer的盐浓度算是有点高的了，
在这样的浓度下还能结合的，应该属于interaction比较强了吧。不知道具体是个什么
蛋白，但是根据自己的经验，如果是这样的话，monomer很可能是没有activity的，必
须要以这样的形式存在才行。你在60kd看到的其他杂带有可能是因为他们本身就是那个
size附近的，或者试试ion exchange来去除杂蛋白？如果一定要用gel filtration，可
以在前面加一步ammonium sulfate precipitation试试，有可能把你的蛋白和杂蛋白分
开一些，但是这个就要摸条件了

的不同而改变。另，对于量子产率
的测量，要求样品与参比的发射峰位近量接近，所以用R6G当quinine sulfate的参比，

我们现在有好几种中药，拿不定，还没换药，仍在吃这副：
生地 玄参，麦冬，五味子 山药、山萸肉，知母，柴胡，赤芍，丹参
我自己以前西医，但更相信中医，看了许多中医，感觉真能用好中医太难，越看越不敢
给妈妈这种疑难症用药。
家里现在还有一方，别人给开的，也没敢吃，怕吃了老人家受不了，不知为何用附子，
生石膏，肉桂，如果她是阴虚火旺，是不是引火归源？

发帖数: 473 | 46 昨天和今天考了step3，但愿pass, 跟usmle说88了。从考完CK到现在，都一年了。当中
经历了跨州搬家，CS, OB, 第二个宝宝出生，申请，面试。
去年6月就买了uw ， 但只做了mcq, 后来肚子太大懒得去考，就搁下了。直到差不多面
完，12-1月才pickup。这次又买了3个月的uw mcq+ccs, 重新做题。MTB是上次看的，这
次没看，但是看了一遍CK first aid, 因为很多基本的东西都忘了。Family practice
看了一半。从考试内容来看，如果说什么东西还占上点边，那就是uw了。还有CK基础很
重要。我可以这么说，pony要是明天就去考step3, 假设她ccs已经练好了，肯定是99，
因为实在没有什么额外的东西。我考的不好是因为基础就不扎实，再让我看上一年也就
这样了，因为脑容量有限，记不住就是记不住。
第一天7个block, 5个office觉得还能handle。那2个 ER, 题目又臭又长，很多看了2遍
也不知所云，勉强做完。前一晚又没睡，考时觉得恶心，然后跳出来的图又增加了恶心

1)比如，我目前复习的儿科的antidose. 对于
anticholinergic的toxic sign， antidose是 Physostigmine sulfate.我期望我能够
大脑里反映的出 anticholinergic + physostigime sulfmate的生理反应机理和药理.
2)比如看congenital adrenal hyperplasia，我就会回去看看adrenal gland 的生理机制
，还有在不同步骤各种酶。这样我就会回去看看病例生理的书。
3)UW中的解释的各个选项，我也会大概的翻翻书了解一下
我这样是否太细了？我也知道越是不熟悉的科弄得越细，熟悉的科目，自己都知道那里
是重点那里掠过。所以时间安排也合理。
关于药物治疗，是否适应UW的那样要求就可以了？
大家帮我提提建议。

一般ck会出一大堆症状体征，首先，你要判断是不是中毒，然后选药物。
2)比如看congenital adrenal hyperplasia，我就会回去看看adrenal gland 的生理机
制，还有在不同步骤各种酶。这样我就会回去看看病例生理的书。
CK一般不考机制，但是考诊断，一大堆体征，症状，似是而非，让你选诊断。
3)UW中的解释的各个选项，我也会大概的翻翻书了解一下
CK就是要教你在无限的模糊的data中找出你的正确诊断。所以，不论什么熟悉不熟悉，
能辨伪存真才是硬道理 |

发帖数: 44 | 50 今天得知了step3的成绩，210+，心里的石头终于落下。终于轮到我给USMLE说再见了。
回顾从step1自己闭门造车考了只带一个9的分数， step2开始参加小组学习，做了所有
能做的题只为拿个99，虽胸有成竹且考试感觉很好，最终但又得到一个9的分数。虽然
这次成绩不高，依然很高兴，毕竟不用发愁如果没过的话住院医开始后没有时间复习该
咋办。过去的3年中得到太多人的帮助 (此处略去人名20个)，绝大多数甚至从未谋面。
今天发贴也是为了回报我们这个社区吧。
自我介绍：毕业>15年，220+/220+/1st/210+；
复习时间：6月，头4个月每天约2-3小时，周末由于陪孩子们玩（前两年欠的太多，你
懂得）及做点家务（再不做但心被老婆炒掉），反而复习不了多少；最后2个月每天4小
时，鼓励自己考了这个2年内再不用受这个罪。
复习资料：随大流，MTB 3, UW CCS, UW step3 Qbank, UW step2 Qbank;
模拟：考前2月，NBME 1 190+, 考前1周，NBME 2 220+;
