loxp, 然后用tamoxifen 去induce， 要求我们提供一个brain specific的gene 去放
Cre -ER
我查了几个常用的brain specific的promoter
BDNF，但是似乎不是brain specific， 因为retina， prostate，kidney 也表达
CaMKII BAC, 这个貌似用的人不多，但是好像有文献说适合brain specific的knockout
不是做这个的，对于KO的知识仅限于Cre LoxP了.
有没有好心人给点建议， 该用那个 gene来放Cre-ER?谢谢！ |

strand specific:
1. strand-specific or not 不影响mapping，因为不管用什么protocol，出来的reads
map到哪条strand都算。再算gene expression的时候才会用到strand information，
具体怎么count reads to gene, 这个要取决于你的strand specific library prep
protocol
2. 拿tophat/bowtie来说，mapping的时候有一个选项--library-type，这个怎么选都
不影响mapping，但是一定要注意，接下来用cuffilink/cuffdiff和htseq-count之类的
，这个--library-type一定要添对了，这个影响gene expression的counting

如果成立一个LLC公司，并且选择specific effective date在2013年12月01日而不是
real filing date，那么在2013年12月01日之前，组织运营这个公司的消费是否可以在
schedule C上面作为business expenses给deduct掉？
那么如果选择2014年1月1日作为specific effective date呢？在federal tax return
方面究竟有什么区别？是不是2013年的任何消费都不能作为deduction了呢？

，让我看看合不合适，因为技术细节她也不清楚，目的就是把job写得非常specific，
非我莫属那种。
有几个问题：
1. HR说她想把position都不写成software engineer相关的，不然会受到一大堆resume
.我当然不反对她如果要改成researcher之类的东西，但是改title会不会对申请绿卡有
影响？毕竟我h1b是申请的software engineer.
2. job description怎么才能尽量specific？说要求在conference上发过paper也可以
吗？要求有phd？还可以说要求懂chinese？还可以说要求在微软工作过不，比如prefer
prior working experience in MSFT什么的...？或者限定工作经验年限恰好在我的区
间内？还可以说要求有在windows phone上publish过app这样行不行？
没搞过，小公司就是麻烦...多谢。 |

所有的One step RT-PCR kit都不可能用gene specific primer来做RT这一步
都是用的oligodT之类的primer来做RT,然后在接着标准的PCR而已
只不过对Taq的要求高一点而已,需要HotStart Taq
user自己加入自己的specific primer for gene of interest.
整个mixture其实就是先RT在PCR, 一般Protocol都要求42度1小时RT,然后直接65赌灭活,再开始PCR的cycle.
我以前用过很多One-step RT PCR,如果不出条带一般是自己的引物的问题
至于One-step realtime PCR 没试过

2 我们想比较这个诊断方法在不同病人身上的表现 我打算计算每个小组的
sensitivity 和 specificity 然后分别有chisq检验 可以吧 因为各小组病人不重叠(
比如 男病人和女病人) 满足indenpendent原则 但是如果病人在不同小组有重叠该怎么
检验呢 比如胖的病人和男的病人 其中必然有些男胖病人 该怎么办
2如果比较不同的诊断方法 1和2 病人都是同一群 该用什么检验方法 mcnemar 比较
paired data 但和这个没关呀
