How about SP6 promoter? what kind of genotype can not express SP6 promoter ?
Can this genotype E. coli K12 ∆(araD-araB)567, ∆lacZ4787(::rrnB-
3), lambda-, rph-1, ∆(rhaD-rhaB)568, hsdR514 express SP6 or T7
promoter-driven gene?

In SP4, there is a new atapi.drv, use that so NT4 can
support HD > 8GB.
I do not know if it is included in SP5 or SP6.
SP3 has sth to support video card.
You can check microsoft
knowledge base, search "large hard driver" etc. then you
should find
some article for the whole installation procedure. I
followed the
procedure, installed sucessfully of NT, 98, Linux. Finally
you should
install sp6. |

进入同样切过的质粒。需要订4个引物，两端两个，中间含有SP6的两个。两端酶切位点
选择片段不长不短，几百个bp合适PCR的就好。

If your are NOT using NTFS for your NT's system dir, try to copy a normal
kernel32.dll and gdi32.dll from other NT4 + SPn (n should be your previous
SP number before SP6) platforms.
RichWin did a very dirty thing here. It renamed the original kernel32.dll to
lernel32.dll and gdi32.dll to hdi32.dll. Then put its own dll wrappers
there.

没有那个OS试图给如此多的硬件软件以及各种流行标准提供支
持，并且基本上作得还可以。BUG是难免的，尤其对稍大系统，
(最近BBS当得比你的PC勤吧:)，但是windows系统的确是在不断
提高。我觉得NT4+SP6个人使用相当的稳定，没有其他那个单机
OS能在提供同样稳定性能的同时还具有如此友好的界面和众多
的应用支持。
当然，现在即使不懂计算机的人也都知道有个Linux要取代windows，
人心向背与迫不及待是明显的。当然调调IRQ，为PCI声卡找找
驱动有时候也是一种乐趣，但不是人人都有这个能力和时间的。
到windows真正被取代可能还有很长的路要走，在此之前还是
享用微软带来的方便吧。 |

Richwin NT几乎为每一个service pack都出了补丁，所有千万不能装原始版，
一定要用补丁程序启动安装过程。所有管理RichwinNT和NT service pacek
的交替升级是很麻烦的事，最好就是别装:) 改用其他中文系统。删除richwin
之后用NT安装盘修补，很多文件的版本可能就乱了，所以不稳。建议安装最新
的service pack， 应该是SP6和SP6a吧。networking设置的很多问题都是旧版
系统文件覆盖了新版造成的。 |

当程序界面出现后, 有1到2分钟的呆滞期, 不响应任何键盘和鼠标输入.

序回来，t7和sp6返回的基本都差不多，就那100bp左右。。我傻了。。。其他2.3kb呢

过没有保真性的酶，而是RNA polymerase II， 如果不用同位素的话，还有什么手段可
以检测不至于敏感度不够高？用RT PCR，模板DNA太多，消化不完全，negative
control都是阳性结果。。。。 |

发帖数: 164 | 39 Did you use denature gel? RNA gel? Normal DNA gels may not give you good
estimation on the size. Purification I used RNAeasy kit from Qiagen, works
well. You could use classic pheno-chloroform extraction if you don't have
the kit. If your RNA coded product has fluorescence or very specific
phenotypes in your system maybe a direct functional verification is easier.

200nt template指的是单链模板么？至少promoter区要双链的。
而PCR产物则是双链的，你的问题会不会是这个原因。
