发帖数: 589 | 4 最近几天用Pierce的Seize X Protein A kit做IP，要沉淀的是一个膜受体。用的抗体
（兔IgG）应该没有问题，因为做WB和flow cytometry都成功。使用了DSS让抗体和
Protein A交联。用了300～400 ug total protein沉淀，4度翻转过夜。之后清洗用的
是自己照着kit说明配的PBS,用kit自带酸性elution buffer洗脱，收集了3个组分。可
是之后用SDS-PAGE+WB检测，受体的带完全没有。我知道这个受体肯定是表达的。请问
可能原因有哪些？
另外Seize X的kit刚好用完，有没有什么其他的kit便宜质量又不错的？曾经考虑过
dynabeads，但是貌似还得另买一台磁力沉淀器？

朗朗在白宫宴请胡锦涛的晚会上选择弹奏电影“上甘岭战役”中的插曲“我的祖国”一
事，这几天在各个中文网站掀起了阵阵波澜。有人称朗朗是一个替共产党唱赞歌的御用
钢琴师；有人指责他有意侮辱美国；还有人怀疑，选这首曲子是否是个人行为？还是与
中国政府的预谋？朗朗和中国政府想要借这个曲子向美国传送什么信息？也有人觉得这
不过是一个无心之举，不必上纲上线。我觉得中国人不必去替美国人操心他们是否受侮
辱，因为显然美国人并不在乎在白宫演奏了抗美援朝的曲子。到底当初选这个曲子是否
有意要侮辱美国现在也不再重要，因为哪怕是有意，显然没有在美国人身上奏效。但是
，朗朗在这一事件后的两个不同回应却让我看到了朗朗没有灵魂不诚实的一面。
朗朗在白宫演出结束后在自己的博客里写到：“能够在众多外宾，尤其是在来自“五湖

颁布，离真正的法治和民主的路还任重道远；等飞到夏威夷了，这也的确是美国，
可是上面除了毛利土著、日裔华裔、海军大兵和旅游者之外，要想看看典型的美国
人恐怕还是找错了地方，这好比Simon De Montfort和Edward I时期议会刚刚试运
转，机制开始建立，模子已经越来越成型了，但还不能算货真价实；只有飞过了落
山鸡，才是真正地到了美国，好比经过了光荣革命，英国以至近代的法治和民主才
