而且一定要狂喝水
现在我一天是300多gram的protein intake （我知道吃爆了，吃那么多没用，主要是
为了保持3000 cal的每日摄入，同时40%来自于蛋白，所以算下来就这么多了）
每天就经常口渴，然后就喝比较多的水。但是睡前不会特意和很多水
以前从来晚上睡觉不起夜的，现在要起来1~2次上厕所
去查了一下专业文献，居然说尿量跟蛋白摄入量无关。。。不知道专家有啥说法没？

just use HPLC to purify them from crude E.Coli expressed protein.
If you don't have one way to do that.
Refolding involves (1) denaturing and reducing (2) suddently dilute
the sample extensively to mild condition (3) get the soluble protein.
Try to search pubmed for methods or refer to any classic protein biochemistry
protocol book. Note: the yield is low(less than 10% of the crude protein
will be correctly refolded). |

the protein you are interested in, say LHRH, has N terminal sequence
identical as protein A. Now you want to use RT-PCR to get the cDNA for
this protein.
I am not sure if protein A is also in your organism, if not, RT-PCR might
work, if yes, RT-PCR won't work as you will get cDNA for protein A...
I personally think you should try two methods, maybe 3 to be sure:
1. RT-PCR.
2. cDNA library
3. Genomic library.
method one is easier to try, but hard to be sure, |

higher NaCl, or wash the pellet afterwards, to decrease the protein-DNA

10kDa to 3kDa) on the gel. I tried to wash my protein several times using
the amicon centrifugal filter at 10kDa cutoff, but I still see those
degraded protein bands on the gels. I also add proteinase inhibitor and also
do the purification on ice to prevent the degradation, but it does not help
. Does anyone know what is the best way to purify or get ride of these
degraded protein bands on the gel? thanks. |

I don't know much about protein-DNA interaction. But the enhancement of
protein's binding with target DNA sequence by phosphorylation should not be
a big surprise.

all proteins, like some membrane proteins, they are really hard to disolve.
High concentration of TritonX-100 will affect SDS-PAGE, you can try 8M urea,
or you can even add 8M urea when casting the SDS-Gel, search methods with key
words like" disolve membrane proteins", "membrane protein SDS-PAGE", you'll
get more informative stuff.

membrane protein) has a low yield which can not be found by comparing
induced and non-induced samples.
If your protein is really important for you, try add a Thrombin, TEV or SUMO
protease cutable His-tag and do a simple separation with Nikel beads. Then
check if there is difference between SDS-PAGEs with and without protease

I first did the expression assay and found protein expression was induced
after adding IPTG by testing the whole cell lysate. (bacteria at certain
