function很快，virus进细胞后表达gRNA/Cas9，马上ko了一个pathway上的基因，3天的
puromycin selection期间，没整合进genome,这个细胞也可能survive的。所以，我以
为sequencing整个genome来找呢。
既然是比较两个pool,遗漏一些或false positive是难免的。估计搞的几个就好。现在
的signal transduction的“网络”已经很成熟了，找到几个估计就会有谱了。看了
Feng zhang的method section,细胞用量很大，估计是不是考虑了整合进genome的效率
不是很高这个因素。
我这个因是新的pathway,自己有potency到了drug级别的small molecule inhibitor，
所以，就想自己试试。

ur molecule need to be lipophilic in order to pass the cell membrane(lipid
bilayer), if it is hydrophilic, then u need active transport. but as what u
said, too lipophilic will decrease solubility and cause PK, protein binding
, etc. problems. it is a balance. in some cases i saw solubility gave in
to potency, then people will try to make salt, or prodrugs to get around the
solubility problem |

一个是ZMP，这个作用的机制是因为它的结构和AMP类似，所以特异性不强
另外一个是在abbott laboratories高通量筛选出来的分子基础上改进的
high potency and specificity，但是目前还没有结构
作用机制研究表明一方面它能激活AMPK，另一方面抑制AMPK的去磷酸化
所以从某种意义上说，它也算是一个inhibitor吧
至于难度，我没有经验
我觉得只要有binding pocket在那，就能用一些SBDD的方法进一步筛选和改进 |

其他serum， 使用的各种成分最要是animal free（目前不是必须要求）。

kids. Also, something important to keep in mind when treating little kids,
prednisolone liquid tastes MUCH better than prednisone, so if you want to
make kids a little happier and spare them the torture, try Orapred :)
3. Your answer is great. Xopenex is much much more expensive than albuterol,
so a lot of hospitals nowadays debate whether they should even keep it on

I have seen seizures from meperidine PCA but never seen a single seizure
from a single or double dose.
75 mg of IV meperidine has an analgesic potency equal to that of 10 mg of IV
morphine or 100 microgram of IV fentanyl. Believe it or not, a lot of
patients claim only demerol is effective to their pain. I used Demerol 75
mg dose a lot but how many of you use morphine 10 mg often?
