而不是常见的tetramer such as G-quartet.

一模型就可以进行，而且现有的核苷类药物怎么说呢已经算是足够好了，木有公司会继
续投入到这个方向上来。

chronic infection理论上病毒不是actively replicate， 只有cccDNA 在肝细胞的保
存。或者说，用了nucleoside analog， 抑制住病毒的大量复制。 几年，十几年后爆
发。 如果有target 受体的药物，这样的药物也是要终身服用吗？ 否则怎么知道病毒
什么时候爆发，爆发的时候这样组织感染新的细胞。
我的问题概况一点就是，在慢性感染的时候，病毒没有大量复制，会不会对肝细胞造成
伤害？ 这个过程跟HIV也类似。但是我看那个免疫系统的检测，HIV 慢性感染的时候
CD4 细胞一直在慢慢减少，知道爆发的时候才迅速减少。我curious的是 这个慢性的过
程，到底是病毒的什么行为造成了伤害？
我的印象是 分化后的肝细胞不会复制自己。如果能够诱导细胞复制，那么ccc DNA就可
能在细胞的复制中丢失，稀释，从而达到乙肝治愈。 |

数都没有相应的结构，第一个药靶GPCR结构也不过07年才做出来，还有10%左右的ion
channel和transporter drugs，大多数的人的靶蛋白也都没有结构，pipeline上怎么有
结构这一环？最多就是一些enzymes或者说kinases的药物，会用结构去辅助设计，怎么
能说大部分都有结构这一环？
不过针对可溶的enzymes，proteases等用结构辅助药物，确实有一些成功的例子，比如
针对HIV reverse transcriptase的non-nucleoside inhibitors，就有收益于结构
develop出来的。 |
