你最好加Caspase inhibitor block apoptosis，再看PI+有没有变化。
很多时候的death是mixed form.
microarray不能说明什么问题，因为所谓的apoptosis相关的基因不是那么绝对的。
另外一个就是看细胞形态，necrosis能看到细胞破裂。

的增多，但是annexinV+/PI-(apoptosis)的没有变化，可是我的microarray结果显示很
多和apoptosis相关的基因上调了。如果说药物作用时间太长导致apoptotic的necrotic
,那我处理4小时也是同样的结果。
请问，会不会apoptosis 在某些细胞里就会引起细胞膜破裂？ |

于两分法，apoptosis vs necrosis。necorsis以前认为不能被调控，不过后来发现在
特定的treatment条件下induce的necroptosis是RIP kinase dependent的。
necroptosis与necrosis的phenotype一样，不过我认为只能算是necrosis中的一种。最
近有个iron-dependent的Ferroptosis发在Cell上，不过我觉得不是特别striking |

已经解释了我所谓“一般”的含义。也是相对于晓东以前工作的水平和影响。
我不是做Necrosis的，只是觉得它的生理作用和已知的凋亡的意义无法相比。也许只在
某些特殊的细胞和条件下，才会有所谓“可调控的”的坏死。另外线粒体的动态变化和
凋亡的关系已经提出过了，我也不认同“又”开辟了Necrosis的一扇大门（如果你指的
是这个方面）。文章做得很漂亮，尤其是这一篇 Mixed Lineage Kinase Domain-like
Protein Mediates Necrosis Signaling Downstream of RIP3 Kinase。
言语有些夸张和不当的地方见谅，以上为个人观点。

在April 10发表的Molecular Cell杂志封面文章中，王晓东实验室描述了MLKL的磷酸化
是如何导致了细胞程序化坏死的分子机制。MLKL的磷酸化使其从单体状态向多聚体状态
的转化。多聚化的MLKL可以结合多种磷脂，从而由细胞质（弥散分布）转移到细胞膜和
细胞器膜上；并在这些膜结构中形成通透性孔道，从而破坏膜的完整性，引起细胞坏死
。这一点有点类似于细胞凋亡中的重要的蛋白Bax和Bak, 在细胞凋亡领域，Bax/Bak在
细胞凋亡过程在线粒体表面聚集，多聚之后形成孔道，从而释放细胞色素C(Cyto C),
诱导细胞凋亡小体，从而激活Caspase3,最后导致细胞凋亡。

心具体的荧光来源 只要采集总信号就行了。比如说有人在circadian gene (per, cyc)
promoter连个luciferase，就能在自由游泳的鱼里看到表达量的周期变化；等等。我站
内msg你一些别的paper吧
后面这个当然存在direct/indirect relation的问题 这些指标
之所以提出来主要是因为容易上量 而不是因为它们能本质性的反映问题，等screen之后
更精细的研究还是需要的。就拿你这个necrosis做例子 筛出来的molecule得看看是通过
溶栓 是通过降低血管内壁细胞脱落，是通过保护neuron还是什么来降低necrosis的
imaging |

最近回去的人的情况，如云老师所说，绝大多数都有2cns，如果只有一篇cns，那基本
上还有>2的子刊。这个不仅仅局限于胖老师经常说的结构生物学，而是所有方向都基本
如此。
当然这些新回去的绝大部分都是千青（有些不是的是因为回去那一年千青还没开始）。
网页上的Title有的是教授，有的是研究员，估计当时还没有统一。但校内一律都是副
教授，也就是相当于这边的AP。我还特地打听了一下，国内的TT制度好像没有Tenured
Associate这一档，清华5年考核通过了就是正经的full professor。
有些老师看不上国内的科研，我就仅列出两位回国后的publication，咱就不说颜宁等
这种Early T的个例了。说两个没太有名气的，还没有评上T的，各位老师可以参考一下
，看看自己有没有别人这么productive。

cell treatment in this situation. As a matter of fact, I have never heard
of any stem cell treatment that has been accepted as a treatment in
situation other than "experiment".
Arthritis of the hip, requiring hip replacement is very common condition. I
doubt her steroid use is the culprit. Long term steroid use can lead to
femur head necrosis, but we are talking about really long term use. |

发帖数: 411 | 50 What you described doesn't sound like avascular necrosis, ligamentous or
labrum pathology is more likely. X-ray won't help you, MRI is the way to go,
plus if you really have avn, it can be seen earlier on MRI that X-ray.
