Dear all, I am in a hurry to decide if it is worth to inject another ES cell
line of a KO mice.
The ES cell line I am using is SV129 cell line ,which carrying agouti coat
colour.
Right now I am having 9 male chimera mice and 1 female chimera mice, all of
the male chimera mice are with very high agouti number indicating a strong
ES cell contribution, and also, as stated above, a distorted gender ratio.

termite inspector在basement里面发现了field mice的踪迹（具体不知道有多严重，
指给LD看了两颗老鼠屎），现在的owner也放置了老鼠夹。但是inspector专门说是mice
不是rat，说在康州rural area，即使是很好，很干净的区，field mice也是到处可见
，他说99%的独立屋都有field mice，因为这种mice个子小，很容易钻进房子，特别是
冬天要进屋取暖。现在的owner养了条狗，看上去很老，看着巨可怜，特脏，拉撒都在
地下室的 unfinished部分。。前后来了几次都是一进去一股味儿。可貌似就我和LD俩
人闻的到。。。请来的 inpector，contractor，agent谁都闻不到。。。邪门了。。。
我和LD都超级怕老鼠的内种，曾经开玩笑要是家里有老鼠绝对俩人一起蹦到桌子上大叫
。。。
inpector建议找个animal control的公司一年来清个几次应该没问题，也不会被我们看
到什么的。

Should I tell my landlord or do something? Even if my landlord will come to

但是地下室木头上有mice啃过的痕迹，
车库和门廊里也分别发现了mice dropping，
两次看房子倒是没看到活动的mice
这个房子两个月没人住了，
在搬进去之前让pest control的人彻底捕杀一遍再把洞封上会有效果吗？
本人实在是太太太害怕老鼠了。
但是好像因为有mice痕迹就放弃房子是不是有点可笑啊。。。。
如果这种问题很普遍可能就咬牙接受了，然后买个insurance啥的。
对了，院子里散放的几块破木头上，还有flower bed的木头下也发现了地下的那种白蚁
，房子里面的结构据说没什么问题，但地下室都装修了，那些梁啊什么的也看不见。
哎，是不是有了后院就要整天deal with这些乱七八糟的小动物和虫子啊，
搞的我都想住回apt或者买个新点的condo算了。 |

？怎么来评估一只老鼠是否是彻底的KO呢？因为发现有一个gene的KO mice，是利用在
其两个exons之间插入另一个不相关的gene，来使其原蛋白丧失功能，研究发现此KO无
任何生理改变，一般行内都会就此得出结论说此蛋白对生物体来说是dispensable的吗
？但是后续有研究又发现，此KO mice体内有好几种此蛋白的变异体（由不同的exons变
换组合而成），而且KO和非KO的变异体种类和数目还略有差别。那这种还可以称之为KO
吗？还可以说此蛋白是dispensable的否？因为关于变异体的来源众说纷纭，有人还发
现有独特的mRNA对应于不同的变异体。所以也无法就确定说变异体就是由此蛋白的gene

并比较?

STOCK Tg(CAG-cre/Esr1*)5Amc/J. 我set up 了5个mating cage ,都是Cre hetero X
WT mice, 交了3个月。子代genotyping后，发现还有Cre transgene 的数目很少，一般
12个老鼠，只有2个带Cre. 比例才1:6,不是理论上后代应该有1：4含Cre 么？
请教各位，这个比例是正常的么，怎么提高子代Cre的数量阿？

功能没有任何损伤，由此得出结论说A在该器官中的作用是dispensable的。
因为这个，联想到自己目前在做的2个蛋白，都有paper发表过说KO mice无异常（也是
跟我做的一样的system），如果由以上提及的paper的思路，那是不是代表我在本
system中对这样的蛋白进行研究也不可能有什么意义了？因为这么说，这些蛋白尽管在
system中的表达量很高，但没有了似乎也不会有什么影响，它的功能是可以被另外的蛋
白取代的。如果我做出了significance，那又会与KO mice的表型相违背，数据无法解
释。现在我很困惑，想听听大家对此有什么意见吗？谢谢。 |

transmission.
关于如何keep这个line,见过一种观点，是最好一直用long range pcr来keep colony，
维持一个完整的选择压力，也就是说如果一直用插入片段的小片段pcr，可能会导致意
外地保留了一些随机事件，在某一代出现飘变
但是long range pcr成本当然要比普通pcr高多了，光酶就贵了n倍——
另一些常见做法就是就选择一个特意的插入片段，甚至neo casette来genotyping
在大多数实验室似乎也work得很好
不知是否有有经验的同学来谈谈？ |

How protein can be expressed with a stop code, which cause a frame shift?
For my personal experience, I used Rosa-Stop-YFP mice from JAX and cannot
see any leakage in Cre negative mice in any lymphocyte populations.
It's a transgene and thus it is possible that the insert allele can move and
incorporate into other promote region. In that case, you just need to
refresh your line and get a new breeder from JAX.
