acid resin conjugated to the resin for head to tail ligation. Your sequence
should be okay, but remember that longer you make it, the harder it will be.
Do not add proline to the sequence, the solid phase synthesis hates proline
and you will likely to fail the elongation after the proline residue.
However, if you like challenge, feel free to do so. The final cycolization

从今年的院士增选最终结果公布以来，各种评论层出不穷，饶毅的“微博炸弹”、施一
公的“意外落选”、“烟草院士”的争议……除去一些幕后因素，最令我感到难以理解
的是化学部新增院士中居然没有一位是有机化学家，几乎全部集中于无机化学和物理化
学，因此也想借此机会发表一下对于中国目前化学生物学发展状况的看法。
今年上海有机所的马大为教授可以说是众望所归，虽然2002年的JACS风波不了了之（
我个人认 为那次“事故”完全是由水平不济、嫉妒心又极强的日本人的“挑衅”引起
的），但是必须要说马老师在多肽类化合物的全合成领域绝对是全国第一人，就更不用
说 那篇氨基酸催化乌尔曼反应的引用率“神文”了，锦上添花的是在院士增选投票进
入第二轮后，马老师以通讯作者身份在Cell上发表了关于筛选、合成小分子细胞自吞噬
的论文，这显然是全国化学领域目前唯一发表在Cell杂志上的文章（要知道，新中国成
立后，第一篇关于大肠杆菌T2转座子的Cell论文由中科院微生物所发表；而第二篇Cell

发帖数: 80 | 11 就是一般的solid phase peptide synthesis。现在一般用fmoc chemistry。十几个，
二十几个aa分段做，然后native chemical ligation连到一起。有半自动/自动的机器
可以用。
这个技术大体上用了很多年了，也算比较成熟。不过很多细节方面有各种这样的问题。
经常是某一个aa上不去（常常与二级结构有关）。有很多小tricks可以试，改溶剂，
coupling agent，solid support，微波等等。这些个factors都有无数个说法，基本凭
经验。
总之就是思路很简单，但具体细节上就是black art。
我觉得如果一千多刀的话，貌似很划算。没啥便宜的方法，生物表达我不大懂，不过一
样不简单。 |

It is a great technology, although it has its own shortcomings:
1. since this technolgy relies on poly A tail for sequencing, RNA without
poly A tails can not be sequenced directly (this can be overcomed by
ligating a polyA tail to all RNA molecules)
2. the length of poly A tail will affect capture of RNA molecules on flow
cells (the longer the better, but too long may affect later sequencing)
3. it can only sequence the very 3' end o |
